Prognostic impact of genetic alterations and methylation classes in meningioma

Anna S. Berghoff et al.

Summary: Meningioma-relevant mutations and DNA methylation clusters were investigated and found to have correlations with patient survival times. TRAF7, KLF4, and TRAKL mutation genotypes were associated with improved prognosis. DNA methylation clusters showed better prognostic discrimination than individual mutations. Molecular profiling could facilitate more precise prognostic assessment and identification of potential targets for personalized therapy in meningioma patients.

Proteogenomic and metabolomic characterization of human glioblastoma

Liang-Bo Wang et al.

Summary: Understanding the molecular pathogenesis of glioblastoma is crucial for improving diagnosis and treatment. Integrated analysis of genomic, proteomic, and metabolomic data on treatment-naive glioblastomas has identified key phosphorylation events and potential targets for specific tumor alterations. Immune subtypes with distinct immune cell types were discovered, and specific histone acetylation patterns were correlated with these subtypes, providing insights for stratification of patients for more effective treatment.

Molecular landscape of IDH-mutant primary astrocytoma Grade IV/glioblastomas

Queenie Hoi-Wing Wong et al.

Summary: Research shows that IDH-mutant primary glioblastomas have a better prognosis and major molecular differences compared to other glioblastomas. Molecular features like G-CIMP status, MGMT promoter methylation, and TP53 mutation can serve as useful prognostic markers.

TERT promoter mutation status is necessary and sufficient to diagnose IDH-wildtype diffuse astrocytic glioma with molecular features of glioblastoma

Kenji Fujimoto et al.

Summary: This study found that in IDH-wildtype lower grade gliomas, the pTERT mutation is a significant predictor of poor prognosis, while both EGFR amplification and PDGFRA gain also play important roles in prognosis.

IDH-wildtype lower-grade diffuse gliomas: the importance of histological grade and molecular assessment for prognostic stratification

Giulia Berzero et al.

Summary: The study finds that IDHwt grade II gliomas have a relatively higher survival rate, while patients meeting the definition of molecular glioblastoma have a shorter survival period. The majority of patients meeting the cIMPACT criteria have isolated TERT promoter mutations, which are not significantly associated with poor outcomes.

Alternative lengthening of telomeres in molecular subgroups of paediatric high-grade glioma

Simone Minasi et al.

Summary: The study found a strong association between H3.3 mutations and ALT mechanism, highlighting different telomere profiles among histone-defined subgroups, suggesting that acquiring the Gly34 mutation on H3.3 might be sufficient to trigger the ALT mechanism.
