Role of Fasciola hepatica Small RNAs in the Interaction With the Mammalian Host

Santiago Fontenla et al.

Summary: MicroRNAs (miRNAs) are important regulators of gene expression. This study investigated the expression patterns of miRNAs during different developmental stages of the liver fluke Fasciola hepatica. The study found variations in miRNA expression across the stages and identified potential target genes. The most highly expressed miRNAs were found to be involved in host immune response and infection establishment and maintenance. Additionally, fragments derived from tRNA processing and vtRNAs were detected, suggesting their involvement in the host-parasite interaction.

Complementary transcriptomic and proteomic analyses reveal the cellular and molecular processes that drive growth and development of Fasciola hepatica in the host liver

Krystyna Cwiklinski et al.

Summary: The migration of immature Fasciola hepatica parasites within the liver is associated with an increase in protein production, expression of signalling pathways, and neoblast proliferation that drive their rapid growth and development. The secretion of a defined set of molecules, particularly cathepsin L peptidases, peptidase inhibitors, saponins, immune regulators, and antioxidants, allow the parasite to negotiate the liver micro-environment, immune attack, and increasing levels of oxidative stress.

Differential expression of microRNAs and tRNA fragments mediate the adaptation of the liver fluke Fasciola gigantica to its intermediate snail and definitive mammalian hosts

Rui-Si Hu et al.

Summary: The study investigated the transcriptional changes of small RNAs during different developmental stages of the tropical liver fluke Fasciola gigantica, revealing important roles of miRNAs and tRFs in parasite development. Furthermore, comparison of miRNA composition between F. gigantica and Fasciola hepatica showed a high level of conservation with some distinct differences, which could be used for species differentiation.

Summary: Fasciolosis, a parasitic disease of agricultural livestock caused by the liver fluke Fasciola hepatica, is traditionally diagnosed using laborious methods with low sensitivity. Enzyme linked immunosorbent assays (ELISA) offer a faster and more sensitive alternative for diagnosis, with recombinant cathepsin L proteins showing promise as effective antigens in detecting liver fluke infection in sheep.

Fasciola hepatica hijacks host macrophage miRNA machinery to modulate early innate immune responses

Nham Tran et al.

Summary: The parasite Fasciola hepatica regulates host innate immune responses by releasing miRNA, especially fhe-miR-125b, which mimics host miRNA to negatively regulate the production of inflammatory cytokines. This manipulation of the miRNA machinery controlling innate cell function is a fundamental mechanism by which worm parasites disarm the early immune responses of their host to ensure successful infection.

Parasitic Helminth-Derived microRNAs and Extracellular Vesicle Cargos as Biomarkers for Helminthic Infections

Summary: Helminth parasites adapt to their complex lifecycles by expressing a unique repertoire of genes at different developmental stages, including the release of microRNAs (miRNAs) and extracellular vesicles (EVs) which aid in host-parasite interactions, immune regulation, and infection processes. These parasite-derived molecules have potential as novel diagnostic tools in the mammalian host circulatory system.
