High-dimensional and single-cell transcriptome analysis of the tumor microenvironment in angioimmunoblastic T cell lymphoma (AITL)

Joshua C. Pritchett et al.

Summary: AITL is an aggressive lymphoid malignancy with a unique tumor microenvironment characterized by a minority population of malignant TFH cells and a diverse infiltrate of immune cells. Non-malignant populations within the TME, such as B cells and CD8+ T cells, are increasingly recognized for their potential role in AITL biology. These non-malignant populations exhibit specific expression patterns that may contribute to the understanding of AITL pathogenesis.

Tet2 deficiency in immune cells exacerbates tumor progression by increasing angiogenesis in a lung cancer model

Yen T. M. Nguyen et al.

Summary: Immune cells with TET2-mutated clonal hematopoiesis exacerbate lung cancer progression by promoting tumor angiogenesis. Inhibition of S100a8/S100a9 can effectively suppress lung cancer growth.

Histone H1 loss drives lymphoma by disrupting 3D chromatin architecture

Nevin Yusufova et al.

Summary: Mutations in histone H1 play a significant role in driving malignant transformation in lymphomas through remodeling of chromatin structure, leading to changes in epigenetic states and de-repression of developmentally silenced genes. This study establishes H1 as a tumor suppressor and highlights its involvement in three-dimensional genome reorganization, ultimately contributing to aggressive lymphomas with increased repopulating potential.

Dynamic regulation of TFH selection during the germinal centre reaction

Julia Merkenschlager et al.

Summary: The germinal centre is a dynamic microenvironment where B cells expressing high-affinity antibody variants are selected for clonal expansion by limiting the number of T follicular helper cells. T follicular helper cells undergo antigen-dependent selection throughout the germinal centre reaction, resulting in differential proliferative expansion and contraction. Competition between T follicular helper cell clones is mediated by the affinity of T cell receptors for peptide-major-histocompatibility-complex ligands.

Affinity-coupled CCL22 promotes positive selection in germinal centres

Bo Liu et al.

Summary: The study reveals a chemokine-mediated mechanism that links the amount of T cell help received by individual B cells recently to their subsequent ability to attract more help. By modulating the expression levels of chemokine CCL22, high-affinity GC B cells are ‘highlighted,’ leading to preferential help from T-FH cells.
