Fluorescence Recovery after Photobleaching in Colloidal Science: Introduction and Application

Aref Abbasi Moud

Summary: FRAP is a method for determining diffusion in material science, which has a substantial influence on the overall qualities of goods in industrial applications. It is a fluorescence-based approach for detecting diffusion and widely used to measure colloidal system heterogeneity and explore diffusion into complicated structures.

What factors determine the number of nonmuscle myosin II in the sarcomeric unit of stress fibers?

Takumi Saito et al.

Summary: This study investigates the characteristics of nonmuscle myosin II (NMII) in actin stress fibers in nonmuscle cells and proposes factors that determine the number of NMII. Experimental results confirm that the number of NMII lies between the functions of bearing cellular tension under static conditions and promptly disintegrating upon forced cell shortening, allowing cells to adaptively respond to mechanical stress.

Time-varying mobility and turnover of actomyosin ring components during cytokinesis in Schizosaccharomyces pombe

Anton Kamnev et al.

Summary: This study investigated the mobility of eight AMR proteins in fission yeast during different stages of mitosis and cytokinesis, identifying three classes of proteins with high, low, and cell cycle-dependent mobile fractions. The F-BAR protein Cdc15 was found to undergo a reduction in mobile fraction after spindle break-down and initiation of AMR contraction, dependent on the septation initiation network. This work provides a novel strategy for estimating cell cycle-dependent mobile protein fractions in cellular structures and offers valuable data for researchers studying cytokinesis.

Determining the domain-level reaction-diffusion properties of an actin-binding protein transgelin-2 within cells

Takumi Saito et al.

Summary: Researchers have described a framework for determining the local physicochemical properties of cellular proteins associated with immobile scaffolds and applied it to transgelin-2, revealing the protein interacts with F-actin via two separate domains in actin stress fibers, determining the chemical properties of each domain and obtaining its pure diffusion properties independent of the association with F-actin. This approach will be valuable in elucidating the signaling mechanism of cellular proteins associated with stress fibers.
