Synthesis of novel thiazolyl hydrazone derivatives as potent dual monoamine oxidase-aromatase inhibitors

Asaf Evrim Evren et al.

Summary: In this study, a new series of 2-thiazolyl hydrazone derivatives were synthesized and evaluated for their inhibitory effects on monoamine oxidase enzymes. Significant results were obtained, with several compounds showing potent and selective inhibition. Molecular docking, molecular dynamics simulations, and QSAR studies were performed to explore the structure-activity relationship. This study represents the first investigation of dual inhibitors on both MAO and aromatase enzymes.

6-Benzyloxyphthalides as selective and reversible monoamine oxidase B inhibitors with antioxidant and anti-neuroinflammatory activities for Parkinson's disease treatment

Qing Song et al.

Summary: A series of 6-benzyloxyphthalides were synthesized as potent inhibitors of monoamine oxidase B (MAO-B) with antioxidant and anti-neuroinflammatory activities. Compound 8f and 14a exhibited excellent selectivity towards MAO-B, moderate antioxidant activity, and in vitro antineuroinflammatory activities. In an in vivo mice model of Parkinson's disease, 8f and 14a showed better effects than safinamide in improving behavioral disorders and restoring dopamine content.

Monoamine oxidases in age-associated diseases: New perspectives for old enzymes

Yohan Santin et al.

Summary: Population aging leads to a higher prevalence of chronic diseases and rising healthcare costs. Cellular senescence and mitochondrial oxidative stress are key processes in age-related diseases. Monoamine oxidases (MAOs) play a role in pathological aging by being upregulated in aged or dysfunctional organs, contributing to the increased degradation of MAO substrates.

Monoamine Oxidase Inhibitors: A Review of Their Anti-Inflammatory Therapeutic Potential and Mechanisms of Action

Mahyar Ostadkarampour et al.

Summary: Chronic inflammatory diseases have a significant impact on patients' quality of life and financial burden. Monoamine oxidase (MAO) inhibitor drugs have been shown to have anti-inflammatory effects, not only in CNS-associated diseases, but also in various non-CNS tissues. Understanding the mechanism(s) of action of MAO inhibitors is critical for their development as anti-inflammatories.
