Design, synthesis, and evaluation of fluoroquinolone derivatives as microRNA-21 small-molecule inhibitors

Summary: In this study, a series of fluoroquinolone derivatives were synthesized and compound A36 was found to be a potent and specific inhibitor of miRNA-21. A36 significantly reduced the expression of miRNA-21 and increased the expression of its target genes in HeLa cells. A36 exhibited antiproliferative activity against multiple tumor cell lines without cytotoxicity in normal cells. Furthermore, A36 induced apoptosis, arrested cells at the G(0)/G(1) phase, and inhibited cell-colony formation in HeLa cells. This study demonstrated that A36 is a novel miRNA-21 inhibitor.

ATF4 promotes lung cancer cell proliferation and invasion partially through regulating Wnt/β-catenin signaling

Summary: The study found that ATF4 expression in non-small cell lung cancer is associated with cancer cell growth and invasion, and it participates in cancer progression by promoting the activity of the Wnt signaling pathway. ICG001 can inhibit the function of ATF4, but when ATF4 and ICG001 are combined, the invasion ability of cancer cells significantly increases, indicating that ATF4's function is not solely dependent on the Wnt/beta-catenin signaling pathway.
