Polycation Architecture Affects Complexation and Delivery of Short Antisense Oligonucleotides: Micelleplexes Outperform Polyplexes

Cation Bulk and pKa Modulate D block Polymer Micelle Binding to pDNA

Summary: Polymer-based gene delivery using polycations relies on the binding, protection, and release of nucleic acids. This study focuses on cationic micelles formed from block polymers and examines their potential as gene compaction agents and carriers. Six amphiphilic block polymers with different cationic groups were synthesized and assembled, resulting in micellar structures with low size dispersities and hydrodynamic radii below 100 nm. Binding of micelles to DNA was influenced by the type of cation present in the micelle corona. This work demonstrates the potential for optimizing and expanding the range of cationic micelle delivery vehicles through simple synthesis and formulation methods.

Molecular Weight Dependence of Block Copolymer Micelle Fragmentation Kinetics

Physical Property Scaling Relationships for Polyelectrolyte Complex Micelles

Summary: Polyelectrolyte complex micelles (PCMs) are self-assembled nanoparticles formed by the associative phase separation of hydrophilic neutral-charged block copolymers and polyelectrolytes, and are effective in delivering hydrophilic payloads. This study investigates the scaling behavior of physical micelle parameters using various experimental techniques and provides specific conclusions, such as the significant increase in PCM core size with the length of the polyelectrolyte block chain, which is crucial for rational PCM design. The results are compared to existing data and theory, showing strong agreement and suggesting the generality of these laws for PCMs.

Cationic Bottlebrush Polymers Outperform Linear Polycation Analogues for pDNA Delivery and Gene Expression

The research demonstrates that bottlebrushes are more efficient in shuttling pDNA into and around the nucleus, highlighting the potential for bottlebrushes as effective biomacromolecule delivery vehicles.
