Cancer progression as a sequence of atavistic reversions

New predictions are made based on evolutionary transitions in our lineage, such as eukaryogenesis and oxidative phosphorylation. Reverting to single-cell biology: The predictions of the atavism theory of cancer

Summary: Cancer initiation and progression may represent a systematic reversion to simpler ancestral phenotypes in response to stress, a concept known as the atavism theory. Research has shown that cancer cells tend to over-express evolutionary older genes and rewire the architecture linking unicellular and multicellular gene networks. Some cancer hallmarks, such as the elevated mutation rate, may be driven by ancient homologs of SOS genes, activated in stressed bacteria for biological workarounds.

USP29-mediated HIF1α stabilization is associated with Sorafenib resistance of hepatocellular carcinoma cells by upregulating glycolysis

Summary: The study identifies USP29 as a regulator of HIF1α and aerobic glycolysis during Sorafenib resistance development in hepatocellular carcinoma. USP29 stabilizes HIF1α transcriptional activity by deubiquitylating, leading to upregulation of glycolytic pathway enzyme HK2, and promoting resistance to Sorafenib in HCC cells. The absence of USP29 restores sensitivity to Sorafenib and correlates with patient response to therapy, suggesting a potential therapeutic target for Sorafenib-resistant HCC.
