The H+-ATPase (V-ATPase): from proton pump to signaling complex in health and disease

Summary: The primary function of H+-ATPase is to create an electrochemical proton gradient across eukaryotic cell membranes to energize cellular processes. It acidifies intracellular vesicles and organelles for essential cell biological events. Additionally, V-ATPases play central roles in normal and pathophysiological processes that impact human health, influencing areas such as cancer, neurodegenerative diseases, and sensory perception.

Purification of active human vacuolar H+-ATPase in native lipid-containing nanodiscs

Summary: V-ATPases are large proton pumps found in virtually every eukaryotic cell, with some isoforms of the a subunit playing a role in tissue-specific diseases and cancer progression. Purification of active human V-ATPases is challenging due to isoform complexity and low abundance, but a strategy involving lipid nanodiscs has been developed for isolation and analysis of the enzyme.
