An approach for mixture testing and prioritization based on common kinetic groups

Albert Braeuning et al.

Summary: With the increasing exposure to chemicals, the issue of potential mixture toxicity has gained attention. However, the current toxicological risk assessments mainly focus on individual compounds, and the approach falls short in addressing cases with dissimilar or independent modes of action. This manuscript suggests considering toxicokinetic effects for grouping substances and proposes common kinetics groups as an additional tool for mixture prioritization.

Immunomodulation and exposure to per- and polyfluoroalkyl substances: an overview of the current evidence from animal and human studies

Evangelia Antoniou et al.

Summary: This article evaluates the association of PFAS with immunomodulation and finds limited evidence from human epidemiology and animal studies to support this association. Lack of association and limited understanding of the importance of antibody levels suggest that more evidence is needed to select immunomodulation as a critical endpoint for PFAS risk assessment in humans.

Application of a Framework for Grouping and Mixtures Toxicity Assessment of PFAS: A Closer Examination of Dose-Additivity Approaches

Philip E. Goodrum et al.

Summary: This article presents a new and systematic analysis of available PFAS toxicity study data, finding that concentration addition and relative potency factor methods are inappropriate, and that the dose-response curves for long-chain perfluoroalkyl sulfonic acids differ significantly from short-chain PFSAs and perfluoroalkyl carboxylic acids. Therefore, it is recommended to conduct a screening level risk assessment by grouping perfluoroalkyl carboxylic acids and PFSAs separately based on the current state of the science.

A review of species differences in the control of, and response to, chemical-induced thyroid hormone perturbations leading to thyroid cancer

John R. Foster et al.

Summary: The review summarizes the current state of knowledge on the physiology and production control of thyroid hormones, as well as the effects of chemicals on their synthesis and release, particularly on thyroid cancer. There are differences highlighted between humans and rodents in thyroid physiology, and ongoing debates about the potential neurodevelopmental consequences of low thyroid hormone levels. The article also emphasizes clear dose-related thresholds for chemical-induced thyroid cancer in rodents, and qualitative and quantitative differences in the transport and metabolism of thyroid hormones between species.

PFAS Exposure Pathways for Humans and Wildlife: A Synthesis of Current Knowledge and Key Gaps in Understanding

Amila O. De Silva et al.

Summary: The understanding of human and wildlife exposures to PFAS has mainly focused on a few traditional substances, but there is a need for assessment of a wider range of new PFAS. Diet is a major pathway of human exposure to some PFAS, but there is significant variability across populations and compounds. Total fluorine data and information on unidentified organofluorine are essential, and water has been identified as a major exposure source in contaminated communities. As water sources are remediated, other sources of exposure such as dust, personal care products, and indoor environments may become more significant.

Systemic PFOS and PFOA exposure and disturbed lipid homeostasis in humans: what do we know and what not?

Styliani S. Fragki et al.

Summary: The association between PFOS/PFOA and increased blood lipids in humans has been observed, while rodent studies show opposite effects, likely due to different exposure levels. Mechanistic research suggests that PFOS/PFOA activate the PPAR alpha pathway in human liver cells, but the involvement of other nuclear receptors is less clear. Future studies focusing on human-relevant test systems are needed to better understand the implications of PFOS/PFOA exposure on lipid homeostasis and human health.

Guidance Document on Scientific criteria for grouping chemicals into assessment groups for human risk assessment of combined exposure to multiple chemicals

Simon John More et al.

Summary: This guidance document offers harmonised and flexible methodologies for grouping chemicals into assessment groups for human risk assessment of combined exposure to multiple chemicals. The framework proposed includes hazard-driven criteria and prioritisation methods to identify low-priority chemicals and reduce the number of chemicals in an assessment group.
