Proteomics characterization of mitochondrial-derived vesicles under oxidative stress

Summary: This study utilized an in vitro MDV budding/reconstitution assay on cardiac mitochondria to identify and characterize the MDV proteome under graded oxidative stress. The results confirmed known cardiac MDV markers and revealed a complete map of the MDV proteome, shedding light on the potential role of MDVs. Proteins vulnerable to oxidative stress directed the cargo loading of MDVs, with certain complexes and metabolic pathway proteins found to be sensitive to antimycin A. Proteins with hyper-reactive cysteine residues and roles in redox and iron metabolism were also identified in the MDV cargo, hinting at their potential as vital biomarkers during mitochondrial stress.
