Ponatinib, Lestaurtinib, and mTOR/PI3K Inhibitors Are Promising Repurposing Candidates against Entamoeba histolytica

Monica M. Kangussu-Marcolino et al.

Summary: Dysentery caused by Entamoeba histolytica affects millions of people annually. The current treatment methods have limitations, leading to the need for new drugs. In this study, researchers screened thousands of compounds and identified 26 potential candidates, including FDA approved drugs and compounds in clinical trials. These candidates showed activity against metronidazole-resistant E. histolytica and had the ability to prevent parasite recurrence after drug removal.

One health therapeutics: Target-Based drug development for cryptosporidiosis and other apicomplexa diseases

Wesley C. Van Voorhis et al.

Summary: The development of bumped-kinase inhibitors (BKIs) for the therapy of One Health parasitic apicomplexan diseases has shown promise in livestock models of cryptosporidiosis, toxoplasmosis, and neosporosis. Further improvements are needed to expand the potential uses of BKIs for the treatment of diseases caused by apicomplexan parasites in animals and humans.

A Curious Case for Development of Kinase Inhibitors as Antigiardiasis Treatments Using Advanced Drug Techniques

Samantha A. Michaels et al.

Summary: Giardiasis, a neglected parasitic disease associated with poverty, poses treatment challenges due to resistance, but targeting essential kinases in Giardia lamblia may offer new therapeutic options. Repurposing kinase inhibitors in drug development could be a cost-effective approach. Innovative pharmacokinetic modeling, imaging technologies, and CRISPR interference could advance the development of more effective therapies for giardiasis based on kinase inhibition.

Efficacy and Tolerability of Quinacrine Monotherapy and Albendazole Plus Chloroquine Combination Therapy in Nitroimidazole-Refractory Giardiasis: A TropNet Study

Andreas Neumayr et al.

Summary: Nitroimidazole-refractory giardiasis is a growing issue among European travelers, with quinacrine showing high effectiveness but a risk of severe neuropsychiatric adverse events, and albendazole plus chloroquine combination therapy having a lower cure rate.
