B-Cell Lymphomas, Version 5.2021 Featured Updates to the NCCN Guidelines

Summary: In the past decade, advancements in understanding the molecular pathogenesis of B-cell non-Hodgkin lymphomas have led to the development of novel targeted therapies, including small molecule inhibitors of select kinases, antibody-drug conjugates, and small molecules targeting various proteins. These therapies, such as anti-CD19 CAR T-cell therapy, have been approved for the treatment of relapsed/refractory diffuse large B-cell lymphoma, and also show promise for follicular lymphoma and mantle cell lymphoma. These developments are highlighted in the NCCN Guidelines for B-Cell Lymphomas.

Prognostic significance of p53, Sox11, and Pax5 co-expression in mantle cell lymphoma

Summary: The study revealed that TP53 mutations in MCL patients have a negative impact on their survival, and patients with triple-positive expression of p53/Sox11/Pax5 have a higher risk. This risk stratification is independent of other prognostic indicators and may help identify high-risk MCL patients.

Automated PD-L1 Scoring for Non-Small Cell Lung Carcinoma Using Open-Source Software

Summary: PD-L1 expression in non-small cell lung cancer is predictive of response to immunotherapy, and automated PD-L1 scoring using QuPath software shows excellent correlation with manual scoring by pathologists. However, automated scoring tends to result in more 1-49% scores compared to manual scoring. Additionally, automated scoring shows high sensitivity but lower specificity at a 1% threshold, and excellent specificity but lower sensitivity at a 50% threshold.
