REPRISAL: mapping lignification dynamics using chemistry, data segmentation, and ratiometric analysis

Oriane Morel et al.

Summary: This article introduces a methodology called REPRISAL for mapping the lignification capacity of plant cell walls in detail. REPRISAL combines three separate approaches, including chemical reporters labeling, automatic segmentation algorithm, and ratiometric analysis, to analyze the distribution and proportions of monolignol reporters in different cell wall zones. The methodology is applied to study the lignification capacity in Arabidopsis, Populus, flax, and maize, as well as the incorporation of a fucose reporter into noncellulosic cell wall polymers.

Fa(c)t checking: How fatty acids shape metabolism and function of macrophages and dendritic cells

Luis Almeida et al.

Summary: Recent studies have advanced our understanding of the role of free fatty acids in shaping the functional properties of macrophages and DCs. Exogenous fatty acids, such as polyunsaturated fatty acids and their oxidation products, can modulate the metabolic properties of these immune cells. Targeted modulation of fatty acid metabolism in myeloid cells may hold promise in therapeutic settings.

Recent Developments and Strategies for Mutually Orthogonal Bioorthogonal Reactions

Yun Hu et al.

Summary: In the past decade, various metal-free bioorthogonal reactions have been developed to achieve simultaneous double-click labeling with minimal competing cross-reactivities, referred to as "mutually orthogonal". Recently, successful triple ligation strategies have also been described. This mini-review discusses the progress in developing orthogonal bioorthogonal reactions and strategies for achieving simultaneous, mutually orthogonal click reactions in one pot.

A unified fluorescence quenching mechanism of tetrazine-based fluorogenic dyes: energy transfer to a dark state

Weijie Chi et al.

Summary: Tetrazine-based fluorogenic probes are powerful tools in bioimaging, biosensing, and medical diagnostics. Understanding the fluorescence quenching mechanism is crucial for enhancing turn-on ratios of these probes. Utilizing the ETDS mechanism, we demonstrated key factors for improving the turn-on ratios of tetrazine-based fluorophores.

Bioorthogonal protein labelling enables the study of antigen processing of citrullinated and carbamylated auto-antigens

Tyrza van Leeuwen et al.

Summary: Proteolysis is essential for various biological processes, especially in the immune system for activating the adaptive immune response. This study introduces a new approach to image antigen proteolysis throughout the processing pathway in an unbiased manner, allowing for the tracking of antigens during degradation while leaving reactive sidechains open to post-translational modifications. The approach was used to study the fate of antigens in rheumatoid arthritis, revealing differences in antigen processing and presentation.
