Nanopore-Based Single-Entity Electrochemistry for the Label-Free Monitoring of Single-Molecule Glycoprotein-Boronate Affinity Interaction and Its Sensing Application

Haoran Tang et al.

Summary: In this study, functionalized-nanopore-based single-entity electrochemistry analysis tools were used to monitor the single-molecule glycoprotein-boronate affinity interaction. The results showed that the equilibrium association constant of this interaction in a confined nanopore was larger than that in an open system, possibly due to enhanced multivalent synergistic binding. Additionally, the method also demonstrated potential application value for the label-free detection of glycoproteins in real samples, paving the way for single-cell analysis.

Biologically Mediated RAFT Polymerization for Electrochemical Sensing of Kinase Activity

Qiong Hu et al.

Summary: The study presents a highly sensitive assay for kinase activity using biologically mediated RAFT polymerization (BMRP) as an efficient amplification strategy. The method utilizes NADH to initiate and sustain the RAFT polymerization, allowing for efficient fragmentation of TCT compounds and amplification of the sensing signal. This BMRP-based approach is biologically friendly, highly efficient, uncomplicated, and low-cost.

Antifouling Aptasensor Based on Self-Assembled Loop-Closed Peptides with Enhanced Stability for CA125 Assay in Complex Biofluids

Min Chen et al.

Summary: A novel ultralow fouling sensing platform was developed based on designed antifouling peptides that could form a stable loop structure, showing excellent antifouling performance, outstanding stability, and satisfactory long-term antifouling capability in complex biofluids. The developed peptide-based aptasensor exhibited a decent response for the analysis of the cancer antigen 125 (CA125), with a wide linear range and a low limit of detection, capable of accurately detecting CA125 in clinical serum samples. This antifouling strategy-based self-assembled peptide with a loop-closed structure provides a potential path for the development of low-fouling biosensors for use in complex biological fluids.

Coenzyme-Mediated Electro-RAFT Polymerization for Amplified Electrochemical Interrogation of Trypsin Activity

Qiong Hu et al.

Summary: A novel signal-on cleavage-based electrochemical biosensor for highly selective interrogation of trypsin activity at ultralow levels is reported, utilizing coenzyme-mediated electro-grafting of ferrocenyl polymers. The constructed trypsin biosensor involves immobilization of peptide substrates, tryptic cleavage, labeling of RAFT agents, and grafting of ferrocenyl polymers mediated by potentiostatic reduction of NAD(+) coenzymes. This strategy is cost-effective and highly efficient for signal amplification, leading to a highly selective and sensitive biosensor with potential applications in diagnostics and therapeutics.
