Characterization of Insulin Dimers by Top-Down Mass Spectrometry

Simon K. Gammelgaard et al.

Summary: High-molecular weight products (HMWP) are critical quality attributes in insulin biopharmaceutical research and development. This study demonstrates the de novo characterization of cross-linked insulin dimers using top-down mass spectrometry workflows, including the efficient cleavage of interchain disulfide bonds and the identification of cross-link connectivities between chains. Various multistage approaches were utilized to identify the chemical composition and cross-link sites within the dimers, revealing specific cross-link patterns in different induced dimers.

Tracking internal and external ions for constrained peptides leads to enhanced sequence coverage and disulfide bond deciphering

Steven Chin et al.

Summary: Top-down characterization of disulfide-rich peptides and proteins faces challenges due to the constrained amino acid backbone, often requiring chemical reduction or enzymatic digestion. In this study, collisional activation was used to confirm disulfide bond connectivity and localize modifications in synthetic therapeutic peptides. Traditional top-down sequencing experiments may overlook internal and external ions produced through secondary fragmentation pathways, which could simplify the experimental process in accurately depicting the disulfide connectivity and sequencing of these biomolecules.

Generating Informative Sequence Tags from Antigen-Binding Regions of Heavily Glycosylated IgA1 Antibodies by Native Top-Down Electron Capture Dissociation

Jean-Francois Greisch et al.

Summary: Immunoglobulins A (IgA), particularly IgA1, are structurally unique antibodies that play a crucial role in defending mucosal surfaces against pathogens. Current methods are unable to separate the millions of unique IgAs potentially present in the human antibody repertoire, making it essential to obtain unambiguous sequence reads of their hypervariable antigen-binding regions for IgA identification. The mass spectrometric method using electron capture dissociation (ECD) shows promise in providing valuable sequence information for the de novo sequencing of IgA1s from milk, saliva, and serum.

Diselenide crosslinks for enhanced and simplified oxidative protein folding

Reem Mousa et al.

Summary: Hirudin, a protein with three disulfide bonds, is commonly used as a model for studying the folding of disulfide-rich proteins. This study investigates the impact of diselenide bridges at different positions on the folding kinetics, yield, and heterogeneity of hirudin, demonstrating enhanced folding rate and yield while reducing complexity. The findings suggest that diselenide substitutions could be a useful tool in designing and characterizing disulfide-rich proteins.

Characterization and Quantification of Selenoprotein P: Challenges to Mass Spectrometry

Jeremy Lamarche et al.

Summary: SELENOP as an emerging marker for selenium nutritional status and various diseases still requires further investigation on its chemical characteristics and improvement in accurate quantitation methods; the presence of multiple isoforms and post-translationally modified variants of SELENOP poses challenges for accurate quantification; mass spectrometry techniques offer many advantages and complementary features in the characterization and quantification of SELENOP and should be carefully evaluated.
