Framework Nucleic Acids in Nuclear Medicine Imaging: Shedding Light on Nano-Bio Interactions

Hao Wang et al.

Summary: Framework nucleic acids (FNAs) are nanoscale oligonucleotide assemblies with unique properties, prepared by following Watson-Crick base-pairing rules. They provide a highly controllable model for nano-bio interactions and allow for analysis of nanostructure-activity relationships. This overview discusses the biological profiles and immune responses of FNAs, as well as recent efforts and challenges in applying them for biomedical purposes. It aims to provide up-to-date information on FNAs and their potential use in selective biomedical applications.

A Small Molecule Strategy for Targeting Cancer Stem Cells in Hypoxic Microenvironments and Preventing Tumorigenesis

Ji Hyeon Kim et al.

Summary: Breast cancer stem cells (BCSCs) are a major contributor to the unfavorable outcomes seen in many breast cancer patients, as they are more prevalent in hypoxic environments and refractory to traditional chemotherapy. A small molecule construct named AzCDF has been developed to target BCSCs effectively in the hypoxic tumor environment, reducing tumor growth and migration. This is the first time a CSC-targeting small molecule has been shown to prevent tumorigenesis in an animal model.

Asymmetric activation of the calcium-sensing receptor homodimer

Yang Gao et al.

Summary: The calcium-sensing receptor (CaSR) is a crucial regulator of calcium homeostasis in humans and a target for the treatment of parathyroid disorders. Research has shown that upon activation, the CaSR homodimer adopts an asymmetric configuration, with one protomer primed for G-protein coupling, stabilized by activator drugs. These findings offer insights for the rational design of therapeutic drugs targeting CaSR.
