An exploratory approach for an oriented development of an untargeted hydrophilic interaction liquid chromatography-mass spectrometry platform for polar metabolites in biological matrices

Summary: This study evaluated different chromatographic columns for analyzing polar metabolites, selecting two new generations of zwitterionic HILIC columns for further evaluation. Using a tree-based method optimization, the study investigated the chromatographic factors affecting retention mechanisms of polar metabolites and demonstrated high coverage of metabolic classes. The optimized methods showed potential for untargeted purposes, with high-quality features ranging from 3,755 to 5,402 for various biological matrices.

Summary: Fatty acyl-coenzyme As (acyl-CoAs) are essential in lipid metabolism pathways, with short-chain acyl-CoAs being part of metabolomics and medium- to (very) long-chain acyl-CoAs being the focus of lipidomics studies. A derivative strategy based on phosphate methylation was proposed in this work, enabling full coverage and good peak shape in liquid chromatography. This method was successfully validated in cultured cells and applied to analyze distinct acyl-CoA profiles in HeLa cells and platelets of coronary artery disease patients.

However, there is a lack of comprehensive analytical method for distinguishing all regioisomeric forms with different acyl side chains and degrees of phosphorylation. This study presents a powerful method for analyzing PIPx profiles in biological samples using a chiral polysaccharide stationary phase and high-resolution mass spectrometry.
