Heparin Enriched-WPI Coating on Ti6Al4V Increases Hydrophilicity and Improves Proliferation and Differentiation of Human Bone Marrow Stromal Cells

Davide Facchetti et al.

Summary: Titanium alloy is a prominent biomaterial for bone contact due to its mechanical loading capability and corrosion resistance. Coating Ti6Al4V substrates with whey protein isolate (WPI) and heparin or tinzaparin improves proliferation and differentiation of bone-forming cells, enhancing hydrophilicity of the material.

Human osteoclast formation and resorptive function on biomineralized collagen

Daniel de Melo Pereira et al.

Summary: Biomineralized collagen composite materials, as an alternative to synthetic bone graft substitutes, offer a biomimetic composition similar to natural bone. In this study, we found phenotypic differences in activity and actin ring formation of human osteoclasts cultured on biomineralized collagen and pure collagen membranes, providing insights for optimized design.

Alkaline Phosphatase Activity of Serum Affects Osteogenic Differentiation Cultures

Sana Ansari et al.

Summary: The study found significant differences in ALP enzymatic activity in FBS from different suppliers, which has important effects on osteogenic differentiation and matrix mineralization. In media with low ALP activity, cellular ALP activity increased and played a major role in mineralization; while in media with high intrinsic ALP activity, less cellular ALP activity was measured, and the ALP activity of the medium also contributed substantially to mineral formation.

Improvement of hydrophilicity and cell attachment of polycaprolactone scaffolds using green synthesized carbon dots

H. Ehtesabi et al.

Summary: The study demonstrated successful surface modification of PCL scaffolds using carbon dots (CDs) solutions to enhance their hydrophilicity, leading to improved cell growth and attachment on the scaffolds. The CD-based scaffold exhibited surface roughness, pore structure, hydrophilicity, and biocompatibility, showing potential for intelligent biomedical scaffolds with adjustable designs and desirable performances.

An Organoid for Woven Bone

Anat Akiva et al.

Summary: This study presents the development of a functional 3D self-organizing co-culture of osteoblasts and osteocytes from human bone marrow stromal cells in vitro, forming an organoid for early stage bone formation. The embedded osteocytes within the collagen matrix produced by osteoblasts mineralized under biological control, showing network formation and communication similar to in vivo osteocytes through expression of sclerostin. The current system represents a comprehensive 3D living in vitro model to investigate osteogenesis in physiological, pathological, and external trigger conditions.

In Vitro Mineralization of Collagen

Bernette Maria Oosterlaken et al.

Summary: Collagen mineralization is a biological process found in various skeletal elements in the animal kingdom, with a focus on understanding bone formation processes and synthesizing tissue engineering scaffolds. This paper reviews in vitro efforts towards collagen mineralization, primarily centered on hydroxyapatite and silica, with a proposed general mechanism for intrafibrillar mineralization of collagen. Future outlooks for improving collagen-based tissue engineering scaffolds are also discussed.

Osteoblast-osteoclast co-cultures: A systematic review and map of available literature

Stefan J. A. Remmers et al.

Summary: Drug research using animal models is costly and time-consuming, with poor translation to clinical trials. To replace and reduce animal models, in vitro cell-culture models like osteoblast-osteoclast co-cultures are being used. However, these studies lack clear identification in titles and abstracts, leading to methodological differences and incomparable results.
