GLP1R Attenuates Sympathetic Response to High Glucose via Carotid Body Inhibition

Audrys G. Pauza et al.

Summary: The study found that GLP1R is expressed in the carotid bodies of rats and humans, and its decreased expression is associated with sympathetic hyperactivity. Targeted administration of GLP1R agonist can reduce the baseline discharge rate of the carotid body and attenuate blood pressure and sympathetic responses induced by chemoreflex. Importantly, GLP1R activation can eliminate hyperglycemia-induced peripheral chemoreflex sensitization and associated sympathetic overactivity.

The role of glia in the physiology and pharmacology of glucagon-like peptide-1: implications for obesity, diabetes, neurodegeneration and glaucoma

Qi N. Cui et al.

Summary: The medical applications of GLP-1R agonists are expanding, with a focus on understanding the impact of GLP-1R signaling on energy balance, glycemic control, neuroinflammation, and oxidative stress in glial cells. The expression of GLP-1Rs in subsets of glial cells and infiltrating macrophages is now supported by evidence, leading to discussions on the effectiveness of GLP-1R agonists for treating metabolic and neurodegenerative diseases, enhancing cognition, and combating substance abuse.

Using a Reporter Mouse to Map Known and Novel Sites of GLP-1 Receptor Expression in Peripheral Tissues of Male Mice

Daniel B. Andersen et al.

Summary: Activation of the glucagon-like peptide-1 receptor (GLP-1R) has various physiological effects in mice, with expression found in multiple tissues including the stomach, intestines, pancreas, kidneys, liver, heart, lungs, and thyroid. This highlights the diverse target cells and effects of GLP-1 and GLP-1R agonists in mice.

Glucagon-like peptide-1 (GLP-1) receptor activation dilates cerebral arterioles, increases cerebral blood flow, and mediates remote (pre)conditioning neuroprotection against ischaemic stroke

Shereen Nizari et al.

Summary: Studies have shown that neuroprotection against ischaemic stroke induced by remote ischaemic conditioning is mediated through GLP-1R signaling, with a potent dilatory effect of GLP-1R activation on cortical arterioles suggesting that the neuroprotection in this model is mediated via modulation of cerebral blood flow and improved brain perfusion.

A survey of the mouse hindbrain in the fed and fasted states using single-nucleus RNA sequencing

Georgina K. C. Dowsett et al.

Summary: This study provides a detailed single-cell level characterization of cells in the AP and NTS expressing receptors for key anti-obesity drugs, revealing their gene expression profiles under different nutritional states. The research also identifies the transcriptional sensitivity of oligodendrocytes to overnight fasting and shows that roughly half of GIPR-expressing cells are oligodendrocytes.

Summary: The study introduces a weighted-nearest neighbor analysis framework to learn the relative utility of each data type in each cell, enabling integrative analysis of multiple modalities. Applied to a CITE-seq dataset, the method constructs a multimodal reference atlas of the circulating immune system and successfully identifies and validates previously unreported lymphoid subpopulations.

Leptin receptor-expressing pericytes mediate access of hypothalamic feeding centers to circulating leptin

Liliia Butiaeva et al.

Summary: Studies have shown that LepR pericytes facilitate localized, paracellular blood-brain barrier leaks, enabling MBH LepR neurons to access circulating leptin. Disruption of LepR in pericytes leads to increased food intake, increased fat mass, and loss of leptin-dependent signaling in nearby LepR neurons.

GIP Receptor Agonism Attenuates GLP-1 Receptor Agonist-Induced Nausea and Emesis in Preclinical Models

Tito Borner et al.

Summary: GIPR signaling blocks emesis and attenuates illness behaviors induced by GLP-1R activation while maintaining reduced food intake, weight loss, and improved glucose tolerance. The hindbrain AP/NTS is crucial for processing GLP-1R ligand-induced effects on food intake and weight suppression, as well as emetic stimuli.

Nutritional regulation of oligodendrocyte differentiation regulates perineuronal net remodeling in the median eminence

Sara Kohnke et al.

Summary: The mediobasal hypothalamus plays an important role in energy balance maintenance, with refeeding triggering proliferation and differentiation of oligodendrocyte progenitors in the median eminence. The remodeling of perineuronal nets in the mediobasal hypothalamus is essential for metabolic functions, and its disruption in obese mice leads to increased food intake and weight gain. These findings highlight the crucial role of oligodendrocyte differentiation in mediating the effects of nutrition on energy balance.

GIPR antagonist antibodies conjugated to GLP-1 peptide are bispecific molecules that decrease weight in obese mice and monkeys

Shu-Chen Lu et al.

Summary: The GIPR-Ab/GLP-1 bispecific molecules show synergistic effects in reducing body weight and improving metabolic parameters, with greater weight loss compared to individual treatments. Additionally, they reduce the respiratory exchange ratio in DIO mice. The simultaneous receptor binding and rapid internalization may explain the efficacy of these bispecific molecules.
