DNA-Damage-Response-Targeting Mitochondria-Activated Multifunctional Prodrug Strategy for Self-Defensive Tumor Therapy

Summary: This study presents a novel multifunctional construct, M1, that specifically targets the DNA damage response in cancer cells. M1 combines a floxuridine (FUDR) and protein phosphatase 2A (PP2A) inhibitor with a GSH-sensitive linker, and is activated in the mitochondria to induce mitochondria-mediated apoptosis. The inhibition of PP2A is highlighted as important for the therapeutic potential in FUDR/5-fluorouracil (5-FU) therapy, and HCT116 xenograft-bearing mice with a low response to 5-FU show significant effects with M1, emphasizing the importance of targeting DNA damage response using tumor-specific microenvironment-activatable systems.

Mitochondria targeted fluorogenic theranostic agents for cancer therapy

Summary: Mitochondria, as the most critical target in cell physiology, have attracted substantial attention in cancer therapy due to their structural and functional discrepancies between normal and cancerous cells. Various synthetic strategies are used to deliver cytotoxins exclusively to mitochondria, achieving significant mitochondrial accumulations by proper selection of cell types and targeting unit. This review article addresses different strategies for targeting cancerous mitochondria with small molecule-based theranostics for diagnostic and potential therapeutic purposes since 2015.

Advances and perspectives in organic sonosensitizers for sonodynamic therapy

Summary: Sonodynamic therapy (SDT) is a promising minimally invasive approach for deep tumor treatment by combining low-intensity ultrasound (US) and a chemical sonosensitizer. Organic sonosensitizers have advantages like defined chemical structure and high ROS generation, but they often face challenges such as poor water solubility and low tumor targeting capacity. Strategies to improve these issues and combine SDT with other cancer treatments are discussed in this review.

A Small Molecule Strategy for Targeting Cancer Stem Cells in Hypoxic Microenvironments and Preventing Tumorigenesis

Summary: Breast cancer stem cells (BCSCs) are a major contributor to the unfavorable outcomes seen in many breast cancer patients, as they are more prevalent in hypoxic environments and refractory to traditional chemotherapy. A small molecule construct named AzCDF has been developed to target BCSCs effectively in the hypoxic tumor environment, reducing tumor growth and migration. This is the first time a CSC-targeting small molecule has been shown to prevent tumorigenesis in an animal model.
