Tumor-associated macrophages in cancer: recent advancements in cancer nanoimmunotherapies

Nisha Kumari et al.

Summary: Cancer immunotherapy has had limited success, but nanoparticles have emerged as crucial tools in targeting tumor-associated macrophages (TAMs) and improving therapy outcomes. Current strategies include restricting TAMs survival, inhibiting TAMs recruitment, and repolarizing tumor-supportive TAMs to an antitumor phenotype.

Microbiotic nanomedicine for tumor-specific chemotherapy-synergized innate/adaptive antitumor immunity

Wencheng Wu et al.

Summary: This study reports a novel microbiotic nanomedicine LOD/TPZ@Lips-LA, which combines lactate oxidase (LOD) and prodrug tirapazamine (TPZ) loaded liposomes anchored on lactobacillus (LA) surface for chemotherapy-synergized antitumor immunity. The nanomedicine induces tumor cell apoptosis through in situ ROS generation and TPZ activation based on LA metabolism, resulting in strong immunogenic cell death (ICD). By regulating biomarker expression, it promotes anti-tumorigenic M1 polarization of innate tumor-associated macrophages and infiltration of adaptive cytotoxic CD8+ T cells, synergistically enhancing the tumor inhibition effects.

Tumor-Microenvironment-Responsive Nanomedicine for Enhanced Cancer Immunotherapy

Shaojun Peng et al.

Summary: Cancer immunotherapy has made great progress in the past decades, but challenges such as low patient response rates and potential immune-related adverse events persist. Tumor-microenvironment (TME)-responsive nanomedicine has shown promise in amplifying immune responses in a specific and localized manner within tumor tissue, leading to increased patient response rates and reduced immune-related side effects.

Macrophage balance fraction determines the degree of immunosuppression and metastatic ability of breast cancer

Chao Wang et al.

Summary: Research suggests that the macrophage balance fraction (MBF) is an effective indicator of the balance of tumor associated macrophages, reflecting the degree of immunosuppression and metastatic ability of breast cancer. Different MBF environments can impact the migration and invasion ability of breast cancer cells, and breast cancer-derived exosomes may play a role in the mechanism of MBF changes in breast cancer.

Ferroptosis-Strengthened Metabolic and Inflammatory Regulation of Tumor-Associated Macrophages Provokes Potent Tumoricidal Activities

Zhengying Gu et al.

Summary: This study demonstrates that an iron-based metal organic framework nanoparticle and a ferroptosis-inducing agent synergistically induce mitochondrial alternation in TAMs, resulting in a radical metabolic switch from mitochondrial oxidative phosphorylation to glycolysis, which is resistant to anti-inflammatory stimuli challenge. The ferroptosis stress strengthened by the nanoformulation also drives multiple pro-inflammatory signaling pathways, enabling macrophage activation with potent tumoricidal activities. The ferroptosis-strengthened macrophage regulation strategy present in this study paves the way for TAM-centered antitumoral treatment to overcome the limitations of conventional methods.

Aspects of the Tumor Microenvironment Involved in Immune Resistance and Drug Resistance

Khalil Khalaf et al.

Summary: The tumor microenvironment (TME) is a complex and dynamic tissue containing various components such as blood supply, immune cells, and extracellular matrix, which together create an environment conducive to tumor cell growth and proliferation. Cellular crosstalk, cell-to-ECM communication, exosomes, and specific metabolic patterns play crucial roles in immune evasion and therapy resistance in the TME. In addition, characteristics like hypoxia, metabolic abnormalities, and mechanical forces further contribute to treatment resistance.
