A synergy between mechanosensitive calcium- and membrane-binding mediates tension-sensing by C2-like domains

Summary: This study investigates the mechanosensitive adsorption of cPLA(2) and its C2 domain to nuclear membranes and artificial lipid bilayers. It is found that stretching the membranes increases the sensitivity of all tested domains to Ca2+ and the binding affinity of cPLA(2)-C2 to stretched membranes is 50 times higher than that to unstretched membranes. These findings provide a quantitative basis for understanding mechanotransduction by C2-like domains.

Summary: Nuclear pore complexes (NPCs) in eukaryotic cells play a crucial role in mediating nucleocytoplasmic exchange. Recent research using cryo-electron tomography and integrative structural modeling reveals that the conformation of NPCs is highly dynamic and can change in response to cellular energy levels and osmotic stress. The data suggests a model where nuclear envelope membrane tension is linked to NPC conformation.

Activation of Piezo1 leads to an increase in [Ca2+](i) through both intracellular release and extracellular influx mechanisms. The enhanced expression and activity of Piezo1 contribute to abnormal elevation of [Ca2+](i) and proliferation in idiopathic PAH-PASMCs.
