Development of a Biodegradable Microcarrier for the Cultivation of Human Adipose Stem Cells (hASCs) with a Defined Xeno- and Serum-Free Medium

Francesco Muoio et al.

Summary: The study developed a novel platform based on chemically defined serum-free cell culture medium and biodegradable microcarrier BR44, enabling the growth of human adipose stem cells while maintaining their undifferentiated state. The BR44 microcarrier allowed hASCs to adhere and grow in a chemically defined serum-free medium, showing better undifferentiated status compared to cells cultured on traditional 2D surfaces.

Long term expansion profile of mesenchymal stromal cells at protein nanosheet-stabilised bioemulsions for next generation cell culture microcarriers

Lihui Peng et al.

Summary: This study demonstrates that bioemulsions formed of microdroplets stabilized by protein nanosheets are well-suited for scale up of adherent stem cells like MSCs. Phenotyping of MSCs cultured on these bioemulsions, solid microcarriers, and classic 2D tissue culture polystyrene over multiple passages shows comparable phenotypes, although stemness is lost at late passages. This validates the use of bioemulsions for long term expansion of adherent stem cells and opens possibilities for novel 3D bioreactors based on microdroplet microcarriers.
