PD-1/PD-L1, Treg-related proteins, and tumour-infiltrating lymphocytes are associated with the development of oral squamous cell carcinoma

Omar Kujan et al.

Summary: This study aimed to evaluate the diagnostic utility of specific immunomodulator proteins and their role in the progression of oral squamous cell carcinoma (OSCC). The results showed that the PD-1/PD-L1 pathway, Treg-related proteins, and tumor infiltrating lymphocytes (TILs) were associated with the development of OSCC. Diagnostic models using these proteins and TILs accurately predicted the grade of oral epithelial disorder.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Beyond immune checkpoint blockade: emerging immunological strategies

Shawn P. Kubli et al.

Summary: The review discusses the history of immunotherapy and checkpoint blockade, emphasizing key clinical failures. It highlights critical aspects of immune processes within the tumor microenvironment, which may guide the development of new therapeutic strategies in immuno-oncology. Future successful therapeutics are likely to focus on rescuing T cell homing and dysfunction in the tumor microenvironment, as well as reappropriating mononuclear phagocyte function for inflammatory remodeling of the TME. New drugs will need to consider the complex cell networks within tumors and across different cancer types.

Parallel genome-wide RNAi screens identify lymphocyte-specific protein tyrosine kinase (LCK) as a targetable vulnerability of cell proliferation and chemoresistance in nasopharyngeal carcinoma

Kitson Liew et al.

Summary: Through genome-wide functional screens, LCK was identified as a key factor in NPC cell proliferation and cisplatin resistance. Depletion of LCK or treatment with LCK inhibitor induced tumor-specific cell death and enhanced cisplatin sensitivity in cells.
