Photocleavable Fluorescent Membrane Tension Probes: Fast Release with Spatiotemporal Control in Inner Leaflets of Plasma Membrane, Nuclear Envelope, and Secretory Pathway

Summary: Mechanosensitive flipper probes, such as PhotoFlippers, containing a photocleavable linker and an ultralong tether, are able to release and label the most ordered membrane accessible by intermembrane transfer upon irradiation. This spatiotemporal control is essential for accessing open, dynamic or elusive membrane motifs without chemical or physical interference. PhotoFlippers have been shown to be useful chemistry tools for studying changes in membrane tension, membrane trafficking, and membrane asymmetry in biological systems.

Thermodynamic and Mechanical Properties of DMPC/Cholesterol Mixed Monolayers at Physiological Conditions

Summary: The study found that cholesterol has various effects on DMPC/Cholesterol monolayers, including fluidization, condensing, and increased stiffness at different concentrations. These effects are more pronounced in pure water at pH around 6.6 compared to buffer at physiological pH = 7.4. All mixtures were found to be thermodynamically stable at specific surface pressures in both pure water and buffer solutions. Additionally, the study compared the stability of these mixtures with a fatty acid monolayer, suggesting that double chain lipids may be better suited for cell membrane formation.

Mechanosensitive channel gating by delipidation

Surface model of the human red blood cell simulating changes in membrane curvature under strain

Summary: This study presents mathematical simulations of red blood cells and their cytoskeleton under linear strain, analyzing changes in curvature and triangular area when the cells are distorted. The highly deformable red blood cells respond to mechanical shape changes with enhanced metabolic flux and cation transport. The reorganization of the simulated cytoskeleton provides insights into the RBC's response to mechanically imposed shape changes.
