Summary: Steric-blocking oligonucleotides (SBOs) are short nucleic acids that modulate gene expression by binding to RNA transcripts and blocking access from cellular machinery. Off-target effects of SBOs were evaluated using RNA-seq, revealing hybridization-driven differential splicing events and other mechanisms driving differential expression events. Machine learning models incorporating splicing predictions improved sensitivity in prioritizing low edit distance hits, but in silico methods are limited in predicting off-target effects of SBOs.
