Notch1 deficiency alters the migratory behavior of developing T cells and calcium signaling in the thymus of medaka

Narges Aghaallaei et al.

Summary: Notch1 signaling plays a crucial role in the proliferation, specification, and selection of developing T cells in the thymus, controlling the migratory behavior of thymocytes and influencing T cell receptor activation. The function of Notch signaling extends to all stages of T cell development in lower vertebrates, except during TCR beta rearrangement.

Phylogeny, Structure, Functions, and Role of AIRE in the Formation of T-Cell Subsets

Daniil Shevyrev et al.

Summary: The most important feature of adaptive immunity is its specificity, which allows for precise recognition of self, altered-self, and non-self. This specificity is based on millions of years of evolution that resulted in the diversification mechanisms of antigen-recognizing receptors and the emergence of the antigen presentation system. AIRE, a noncanonical transcription factor, plays a crucial role in this system by controlling differentiation and expression of peripheral tissue-specific antigens, as well as increasing the diversity and transfer of presented antigens. This review highlights the importance of the antigen presentation system and discusses the role of AIRE.

The evolution and functional characterization of CXC chemokines and receptors in lamprey

Zhaosheng Sun et al.

Summary: Chemokines are soluble peptides that guide cell migration in development and immune defense. The CXC chemokine system in lamprey has been found to have diversified, showing variations in gene organization and expression patterns in different tissues, indicating an evolutionary adaptation in jawless fish.

Immunoglobulin T genes in Actinopterygii

Summary: In teleost fishes, three immunoglobulin isotypes have been identified, with IgT being specific to them. Recent analysis of fish genomes revealed previously undescribed IgT isoforms. Evolutionary analysis indicates that IgT predates IgM in Actinopterygii.

A mechanosensitive peri-arteriolar niche for osteogenesis and lymphopoiesis

Bo Shen et al.

Summary: Stromal cells in adult bone marrow expressing leptin receptor are a critical source of growth factors for haematopoietic stem cells. Among these cells, those expressing osteogenic growth factor osteolectin are poised for osteogenesis, while those near sinusoids are primed for adipogenesis. Mechanical stimulation is crucial for the maintenance of the peri-arteriolar niche for osteogenesis and lymphopoiesis.

Evolution of thymopoietic microenvironments

Ryo Morimoto et al.

Summary: In vertebrates, the thymus plays a crucial role in the development of mature lymphocytes, with the microenvironment of the thymus influencing lymphopoietic activity. The Foxn1/4 family of transcription factors control the formation of a functional thymic microenvironment. Throughout evolution, the thymus transitioned from supporting both B cell and T cell development to specializing in T cell development.

Mucosal immunoglobulins of teleost fish: A decade of advances

Irene Salinas et al.

Summary: Immunoglobulins play critical roles in the immune systems of jawed vertebrates, with IgT identified as playing a predominant role in fish mucosal immunity. Recent studies have highlighted the importance of secretory Igs in maintaining microbiota homeostasis and controlling pathogens at mucosal sites. This review aims to summarize the current status of fish Igs research since the discovery of IgT, while identifying future research directions.

Do ectothermic vertebrates have a home in which to affinity mature their antibody responses?

A. Muthupandian et al.

Summary: This study discusses the evidence for antibody affinity maturation and somatic hypermutation in ectotherms, suggesting that fish melanomacrophage clusters may serve as functional sites analogous to germinal centers. Further research in zebrafish can help validate these predictions, with implications for vaccine use in aquaculture and antibody affinity maturation processes in all ectothermic vertebrates.

Summary: This article presents an overview of the positioning of HSCs in the bone marrow and how their function is maintained, as well as discusses the regulatory role of niches in HSC function and hematopoiesis. Some important unresolved questions in this area have also been identified. The niches play a crucial role in maintaining HSC function and hematopoiesis.

Immunological memory in teleost fish

Michal Stosik et al.

Summary: Immunological memory plays a key role in adaptive immunity, leading to a faster response upon re-exposure to the same antigen. Genetic alterations may maintain immune activity at a low level independently of the antigen. Studies in Teleostei suggest the presence of immunological memory determined by T and B cells, with potential involvement of macrophages and NK-like cells.

αβ/γδ T cell lineage outcome is regulated by intrathymic cell localization and environmental signals

Narges Aghaallaei et al.

Summary: The study in medaka fish reveals that progenitors first enter a thymic niche favoring the development of gamma delta T cells, and the gradients of interleukin-7 and Ccl25a in the niche influence the lineage outcome. This previously unknown mechanism determines the proportion of alpha beta/gamma delta lineages within species.
