The CD40-CD40L Dyad as Immunotherapeutic Target in Cardiovascular Disease

Laura A. Bosmans et al.

Summary: Chronic inflammation drives the development of atherosclerosis, for which additional anti-inflammatory therapeutic strategies are required despite optimal treatment of classical cardiovascular risk factors. The CD40-CD40L dyad plays a central role in regulating the inflammatory response during atherosclerosis development, and novel CD40-CD40L targeting strategies show therapeutic potential in cardiovascular medicine.

Head-to-head comparison of DFO* and DFO chelators: selection of the best candidate for clinical89Zr-immuno-PET

Marion Chomet et al.

Summary: Novel analogues DFO*-NCS and DFOSq showed superior stability compared to traditional DFO in in vitro and in vivo experiments. Conjugates with DFO* exhibited lower bone uptake and provided superior detection of tumor-specific signal in a bone metastasis model. DFO* proves to be a more stable and effective candidate for preclinical and clinical (89)Zr-immuno-PET.

The Role of 89Zr-Immuno-PET in Navigating and Derisking the Development of Biopharmaceuticals

Guus A. M. S. van Dongen et al.

Summary: The identification of molecular drivers of disease and the rise of biotherapeutics have impacted clinical care, but also presented challenges. The application of Zr-89-immuno-PET in drug development shows promise in guiding the design, development, and application of biologic drugs, with increasing potential for understanding drug-target combinations in clinical studies.

Cell-specific and divergent roles of the CD40L-CD40 axis in atherosclerotic vascular disease

Michael Lacy et al.

Summary: The CD40L-CD40 signaling axis plays a role in atherosclerosis. This study investigates the cell-specific functions of the most relevant CD40L-expressing cell types. Deficiency of T cell-derived CD40L reduces and stabilizes plaques through impaired Th1 polarization, while platelet-derived CD40L ameliorates atherothrombosis.

Imaging atherosclerotic plaques by targeting Galectin-3 and activated macrophages using (89Zr)-DFO-Galectin3-F(ab')2 mAb

Zohreh Varasteh et al.

Summary: The study found that targeting Gal3 through PET/CT imaging can effectively track atherosclerotic plaques in mice. The results suggest that Gal3 is mainly expressed in macrophages within plaques, and exhibits similar expression patterns in human plaques.
