Two-dimensional shear wave elastography predicts survival in advanced chronic liver disease

Jonel Trebicka et al.

Summary: Liver stiffness measurement is a useful tool for predicting mortality and development of complications in patients with advanced chronic liver disease. The combination of MELD score and LSM can effectively stratify patients based on their risk levels.

Clinical, histological and molecular profiling of different stages of alcohol-related liver disease

Meritxell Ventura-Cots et al.

Summary: Despite comparable alcohol intake, AH patients presented with worse liver function compared with ndALD patients. Bilirubinostasis, severe fibrosis and ductular reaction were prominent features of AH. AH patients exhibited a more profound deregulation of gene expression compared with ndALD patients.

Lipoprotein Z, a hepatotoxic lipoprotein, predicts outcome in alcohol-associated hepatitis

Kunpeng Hu et al.

Summary: The study revealed significantly elevated levels of LP-Z in AH patients, with a Z-index <= 0.6 predicting 90-day survival. RNA sequencing showed an inverse association between enzymes responsible for VLDL to LDL conversion and AH severity, while altered FC levels in liver explants of AH patients indicated a pathogenic role of FC in LP-Z particles in hepatocyte toxicity.

A genetic risk score and diabetes predict development of alcohol-related cirrhosis in drinkers

John B. Whitfield et al.

Summary: This study developed a genetic risk score that can identify individuals at high risk of developing cirrhosis. It was found that the risk of cirrhosis is increased over 10-fold in patients with diabetes and a high genetic risk score. This risk assessment could lead to early and personalized management of the disease in high-risk patients.

Digital pathology: accurate technique for quantitative assessment of histological features in metabolic-associated fatty liver disease

David Marti-Aguado et al.

Summary: Histological evaluation of MAFLD biopsies is subjective and with interobserver variability. This study found that digital pathology can provide an automated, precise, and objective assessment of histological features such as fibrosis, steatosis, and iron in MAFLD. The digital analysis measurements showed good concordance with pathologists' scores, except for inflammation assessment.

Mortality in biopsy-confirmed nonalcoholic fatty liver disease: results from a nationwide cohort

Tracey G. Simon et al.

Summary: Population-based data in Sweden showed that all histological stages of NAFLD were linked with significantly increased overall mortality, and the risk escalated progressively with worsening histology. Excess mortality was mainly attributed to extrahepatic cancer and cirrhosis, while cardiovascular disease and HCC made modest contributions.

Development and prognostic relevance of a histologic grading and staging system for alcohol-related liver disease

Carolin Lackner et al.

Summary: The SALVE Histopathology Group developed and validated a grading and staging system for alcohol-related liver disease (ALD) and evaluated its prognostic utility in a multinational cohort of 445 patients. The system showed significant associations with disease activity and fibrosis stage, impacting both long-term and short-term outcomes in ALD patients.

Serum transferrin as a biomarker of hepatocyte nuclear factor 4 alpha activity and hepatocyte function in liver diseases

Nurdan Guldiken et al.

Summary: The study found that serum transferrin levels correlated with hepatic transferrin expression in patients with advanced liver disease. Reduced HNF4 alpha and transferrin protein levels were confirmed in individuals with cirrhosis. In alcoholic hepatitis patients, a relationship between transferrin and HNF4 alpha was observed, while inflammatory factors were negatively associated with transferrin signature.

Interaction between metabolic syndrome and alcohol consumption, risk factors of liver fibrosis: A population-based study

Elisa Pose et al.

Summary: Alcohol and metabolic syndrome are common cofactors of liver disease. This study found that metabolic syndrome increases the risk of liver fibrosis in individuals with alcohol consumption, especially in high-risk drinkers. Factors such as male gender, liver enzymes, years of consumption, and metabolic syndrome were associated with liver fibrosis in subjects with alcohol consumption.

Prognostic performance of 7 biomarkers compared to liver biopsy in early alcohol-related liver disease

Ditlev Nytoft Rasmussen et al.

Summary: Alcohol is the most common cause of liver-related mortality and morbidity. This study aimed to evaluate and compare the prognostic performance of elastography and blood-based markers in predicting liver-related events, severe infection, and all-cause mortality in patients with a history of excess drinking. The results showed that transient elastography, enhanced liver fibrosis test, and 2D shear-wave elastography are highly accurate prognostic markers in patients with alcohol-related liver disease.

Genetic predisposition similarities between NASH and ASH: Identification of new therapeutic targets

Cristiana Bianco et al.

Summary: Fatty liver disease can be caused by a combination of factors such as excess alcohol consumption, dysmetabolism, and environmental cues, leading to steatohepatitis and potentially severe liver conditions like liver failure and hepatocellular carcinoma. Recent genetic discoveries have shed light on the causes of steatohepatitis, opening up new possibilities for more targeted risk assessment and potential therapeutic developments. This knowledge could pave the way for precision medicine approaches tailored to individuals at higher risk of liver diseases.
