Protein Secondary Structure Affects Glycan Clustering in Native Mass Spectrometry

Hao Yan et al.

Summary: Research indicates that the determination of specific glycan binding affinities from native mass spectrometry (MS) must be interpreted cautiously, as the degree of glycan clustering during electrospray ionization depends on the physicochemical properties of the protein studied. The mass fraction of beta-sheets in the protein has a strong influence on the glycan clustering, highlighting the importance of selecting a suitable reference protein for analysis.

FoldAffinity: binding affinities from nDSF experiments

Stephan Niebling et al.

Summary: The study evaluates binding affinity quantification based on thermal shifts, finding that results from isothermal analysis are more consistent with established biophysical techniques compared to apparent K(d)s obtained from melting temperatures. Additionally, a method to optionally fit the heat capacity change upon unfolding during isothermal analysis is described, along with the release of a web server for easy application of isothermal analysis to nDSF data.
