A midgut-specific lytic polysaccharide monooxygenase of Locusta migratoria is indispensable for the deconstruction of the peritrophic matrix

Ming-Bo Qu et al.

Summary: Lytic polysaccharide monooxygenases (LPMOs) are enzymes that play an important role in the hydrolysis of recalcitrant polysaccharides in insects. In this study, the midgut-specific LPMO15s were found to be widely distributed in different insect orders. The function of LmLPMO15-3 in Locusta migratoria was investigated, and its downregulation severely affected survival and led to incomplete digestion of the peritrophic matrix (PM) during molting.

Functional and Structural Characterizations of Lytic Polysaccharide Monooxygenase, Which Cooperates Synergistically with Cellulases, from Ceriporiopsis subvermispora

Huyen Nguyen et al.

Summary: Research on a lignocellulose-degrading enzyme called CsLPMO9 has shown that it significantly enhances cellulose degradation efficiency when treated together with a cellulase cocktail. Analysis of the functional and structural properties of CsLPMO9 revealed key residues involved in binding with cellulose substrates, potentially increasing its affinity and promoting efficient biomass utilization.

Characterization of a lytic polysaccharide monooxygenase from Aspergillus fumigatus shows functional variation among family AA11 fungal LPMOs

Fredrik Gjerstad Stopamo et al.

Summary: The study reports the structural and functional characteristics of a single-domain AA11 LPMO from Aspergillus fumigatus. The LPMO, AfAA11A, shows substrate-binding surface features similar to those of known chitin-active LPMOs and exhibits considerable affinity for both alpha-chitin and beta-chitin, enhancing chitin degradation.

On the roles of AA15 lytic polysaccharide monooxygenases derived from the termite Coptotermes gestroi

Joao Paulo L. Franco Cairo et al.

Summary: Lytic polysaccharide monooxygenases (LPMOs) are copper-dependent enzymes that catalyze the oxidative cleavage of polysaccharides. Recent research on AA15 LPMOs from lower termite Coptotermes gestroi revealed that they can oxidize chitin but not cellulose, indicating potential differences in biochemical functions compared to other insects. Structural analysis showed key differences in catalytic site residues of CgAA15a/b compared to other LPMOs, suggesting they may have evolved unique substrate specificities.

The lytic polysaccharide monooxygenase CbpD promotes Pseudomonas aeruginosa virulence in systemic infection

Fatemeh Askarian et al.

Summary: The Pseudomonas aeruginosa lytic polysaccharide monooxygenase CbpD is a chitin-oxidizing virulence factor that promotes bacterial survival in human blood, with its catalytic activity promoted by azurin and pyocyanin. Deletion of CbpD hinders the bacterium's ability to establish lethal systemic infection and leads to increased bacterial clearance. CbpD's key role in virulence is attributed to its ability to attenuate the terminal complement cascade in human serum.

Secreted pectin monooxygenases drive plant infection by pathogenic oomycetes

Federico Sabbadin et al.

Summary: The discovery of copper-dependent lytic polysaccharide monooxygenases (LPMOs) in plant pathogenic oomycetes, particularly Phytophthora infestans, has revealed their crucial role in early infection stages by oxidatively cleaving pectin in the plant cell wall. Silencing LPMO-encoding genes inhibits potato infection by P. infestans, demonstrating their significance in host penetration. Identifying LPMOs as virulence factors in pathogenic oomycetes presents new opportunities in crop protection and food security.
