Antivirals against (Re)emerging Flaviviruses: Should We Target the Virus or the Host?

Maria-Jesus Perez-Perez et al.

Summary: The COVID pandemic has shown our vulnerability and the limited treatment options for emerging infectious diseases. Flaviviruses, transmitted by arthropods, pose a global health challenge with no approved drugs for treatment. In this Viewpoint, the advantages and disadvantages of virus-targeting antivirals and host-targeting drugs are analyzed, highlighting their complementary nature.

Antivirals against (Re)emerging Flaviviruses: Should We Target the Virus or the Host?

Maria-Jesus Perez-Perez et al.

Summary: This viewpoint discusses the advantages and disadvantages of virus-targeting antivirals and host-targeting drugs as potential therapies for Flavivirus infections. These two approaches may be complementary, each with its own strengths and weaknesses.

The structural basis of herpesvirus entry

Sarah A. Connolly et al.

Summary: Herpesviruses are enveloped, double-stranded DNA viruses that establish lifelong infections and cause a variety of diseases. The entry into host cells involves the binding of the virus to specific receptors and the coordinated action of multiple viral entry glycoproteins. While the core fusion machinery is conserved among all herpesviruses, each species uses distinct receptors and receptor-binding glycoproteins.

Effects of epigallocatechin-3-gallate and acyclovir on herpes simplex virus type 1 infection in oral epithelial cells

Ching-Yi Wu et al.

Summary: The combined effects of EGCG and ACV show additive effects on reducing cytotoxicity, viral yields, and viral protein expression during HSV-1 infection in oral epithelial cells. Additionally, EGCG reduces viral particles and DNA levels during viral entry phase, while ACV can reduce the activation of c-Jun N-terminal kinase induced by HSV-1.

Chemical compositions and experimental and computational modeling activity of sea cucumber Holothuria parva ethanolic extract against herpes simplex virus type 1

Mohsen Keshavarz et al.

Summary: The ethanolic extract of Holothuria parva demonstrates significant antiviral activities against HSV-1 by reducing virus titers and interfering with virus attachment to cell receptors. Octadecanoic acid 2-hydroxy-1-(hydroxymethyl) ethyl ester and 2',6'-acetoxylidide are major constituents in the extract. Computational docking analysis suggests that the extract compounds may inhibit virus attachment by covering both virus and host cell binding domains.

Current Drugs to Treat Infections with Herpes Simplex Viruses-1 and-2

Lauren A. Sadowski et al.

Summary: Herpes simplex viruses-1 and -2 can cause lifelong infections, especially posing a greater threat to immunocompromised patients, hence the need for interventions. Experimental vaccines are still under consideration, and current treatment relies solely on antiviral medications.

Rational Design of Live-Attenuated Vaccines against Herpes Simplex Viruses

Brent A. Stanfield et al.

Summary: Diseases caused by human herpes simplex virus types 1 and 2 affect millions worldwide, but there are currently no FDA-approved vaccines. The development of novel live-attenuated vaccines aims to prevent or reduce the impact of herpes simplex infections, building on the success of past live-attenuated vaccines in combating viral diseases. These vaccines are designed to generate robust and protective immune responses based on our understanding of the immunopathogenesis of herpesviral infections.

Research Progress on the Antiviral Activity of Glycyrrhizin and its Derivatives in Liquorice

Changchao Huan et al.

Summary: Liquorice, a traditional medicine, contains triterpenoids such as glycyrrhizin and glycyrrhetinic acid which have inhibitory effects on various viruses, including SARS-CoV-2. These compounds work through mechanisms such as inhibition of virus replication, direct inactivation of viruses, and inhibition of inflammation. Further research on unknown antiviral mechanisms, structural modifications, and drug combinations is emphasized for future studies.
