Anticancer and antileishmanial in vitro activity of gold(I) complexes with 1,3,4-oxadiazole-2(3H)-thione ligands derived from δ-D-gluconolactone

Andres Villasenor Espinosa et al.

Summary: Four gold(I) complexes were synthesized as potential anticancer agents. Increasing the lipophilicity of the ligands proved useful in enhancing their anticancer and antileishmanial properties. The novel compounds showed low micromolar IC(50) values and selectivity against specific cell lines, indicating their potential in cancer and leishmaniasis chemotherapy research.

Anticancer and antibacterial properties of trinuclear Cu(I), Ag(I) and Au(I) macrocyclic NHC/urea complexes

Christian H. G. Jakob et al.

Summary: In this study, the synthesis and characterization of the first macrocyclic Cu(I), Ag(I), and Au(I) tricarbene/urea N-heterocyclic carbene (NHC) complexes were described. The newly described silver and gold complexes exhibited luminescence with emission maxima at specific wavelengths, and showed moderate activity in inhibiting the growth of bacteria and cancer cells. The type of metal in the complexes had a significant influence on their antimicrobial and antiproliferative activities.

Reactivity of antitumor coinage metal-based N-heterocyclic carbene complexes with cysteine and selenocysteine protein sites

Iogann Tolbatov et al.

Summary: The study investigated the reactivity of antitumor metal complexes with protein binding sites cysteine and selenocysteine, revealing a higher affinity towards the deprotonated forms. The neutral forms' binding process was found to be slow, while protonation from an external source was required for the deprotonated forms. The reactivity of all coinage metals towards cysteine and selenocysteine was similar, with Au(I) complex exhibiting higher reaction and activation free energies compared to Cu(I) and Ag(I).

Summary: Long-term survivors of GBM are at risk of developing second primary neoplasms, including leukemia. Classic TKIs are discouraged due to their potential to increase tumor and stem cell migration. The study developed compounds inhibiting tubulin polymerization, showing promising efficacy in both GBM and CML cells, providing a new treatment option for patients.

N-Heterocyclic Carbene-Gold(I) Complexes Targeting Actin Polymerization

Domenico Iacopetta et al.

Summary: Transition metal complexes, particularly NHC-gold complexes, are being extensively studied for their wide range of biological targets, including their role in regulating cell cytoskeleton organization. Two such complexes, AuL4 and AuL7, have been shown to inhibit actin polymerization, contributing to the development of more effective and safer cancer treatments.

In Vitro Nephrotoxicity Studies of Established and Experimental Platinum-Based Compounds

Sarah Schoch et al.

Summary: PN149 exhibits higher cytotoxicity in normal human kidney tubule epithelia compared to established platinum chemotherapeutics like cisplatin, carboplatin, and oxaliplatin. Gene expression profiling reveals induction of ROS-inducible and anti-oxidative genes by PN149, indicating an oxidative stress response. Unlike cisplatin, no pro-inflammatory response was observed with PN149, suggesting reduced nephrotoxic potential.

Summary: Breast cancer is one of the leading causes of cancer-related deaths among women globally, with tumor metastasis and the presence of CSCs contributing significantly to high mortality rates. Research needs to focus on discovering effective anticancer agents that can inhibit the proliferation of differentiated tumor cells and reduce the tumorigenic capability of CSCs.

Platinum-based chemotherapy via nanocarriers and co-delivery of multiple drugs

Caihong Xian et al.

Summary: Platinum-based anticancer drugs inhibit cancer cell growth by disrupting DNA replication, making them widely used in clinics. However, their successful application is limited due to drug resistance and side effects. Strategies such as nanocarriers and multicomponent chemotherapy have been used to address these challenges.

Abdulqader M. Abdulqader et al.

Summary: This theoretical study investigates the preparation of new gold complexes as anti-tumor drug compounds using E-Docking energy between the complexes and protein receptors in the human body. The study identified specific complexes with high binding to the Ras protein, showing potent inhibitory effects on breast cancer cell growth. The results demonstrate that complexes containing curcumin are the most effective anti-cancer agents.
