Chain release mechanisms in polyketide and non-ribosomal peptide biosynthesis

Rory F. Little et al.

Summary: This review discusses the diversity of enzymes and mechanisms involved in the release of polyketide and non-ribosomal peptide natural products, and how they impact the structure of the products. It also explores how these mechanisms can be utilized to introduce structural diversity in engineered biosynthetic pathways.

Transacylation Kinetics in Fatty Acid and Polyketide Synthases and its Sensitivity to Point Mutations**

Franziska Stegemann et al.

Summary: This study investigates the mechanisms of FASs and PKSs in synthesizing important products and the substrate selectivity and enzymatic properties of different ATs. Findings reveal that ATs in PKS systems occurring in bacteria are slower than ATs in mammalian FASs, reflecting the respective purposes and metabolic contexts within the organism.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.

Mapping the biosynthetic pathway of a hybrid polyketide-nonribosomal peptide in a metazoan

Likui Feng et al.

Summary: The authors identify the biosynthetic roles of individual megasynthetase domains and additional enzymes required for nemamide biosynthesis in the canal-associated neurons of C. elegans. They also discover a trafficking mechanism involving at least five additional enzymes for transferring intermediates between a PKS and an NRPS system. This mechanism expands the biosynthetic potential of the PKS-NRPS system and is likely important for the regulation of nemamide biosynthesis.

Mining and unearthing hidden biosynthetic potential

Kirstin Scherlach et al.

Summary: Genetically encoded small molecules play key roles in ecological interactions and drug discovery, but are often difficult to detect due to low production and high rediscovery rates. Genome mining for biosynthetic gene clusters can help address these limitations, leading to the discovery of previously overlooked natural products.

Computational studies on the substrate specificity of an acyltransferase domain from salinomycin polyketide synthase

Huining Ji et al.

Summary: Polyketides are a diverse group of natural products synthesized by modular polyketide synthases (PKSs) from various CoA thioesters. This study investigated the substrate specificity of SalAT14 using MD simulations and QM/MM calculations, revealing a preference for EMCoA. Nucleophilic attack on the thioester carbonyl group was found to be the rate-limiting step in transacylation.
