Cloning and functional analysis of the novel rice blast resistance gene Pi65 in japonica rice

Lili Wang et al.

Summary: Pi65, a novel rice blast disease resistance gene, was cloned from Oryza sativa japonica and can enhance rice blast resistance through genetic engineering. The protein encoded by Pi65 has specific domains that can recognize and resist rice blast fungus. After pathogen infection, Pi65 activates numerous disease resistance-related genes while simultaneously suppressing photosynthesis and carbohydrate metabolism-related genes.

First report of barley blast caused by Magnaporthe oryzae pathotype Triticum (MoT) in Bangladesh

Krishna Kanta Roy et al.

Summary: In April 2019, distinct partially or fully bleached heads were observed in barley fields in Jashore and Meherpur in Bangladesh, indicating the first occurrence of barley blast caused by Magnaporthe oryzae pathotype Triticum (MoT). The fungus isolated from diseased spikes was identified as Magnaporthe oryzae based on conidial morphology, and MoT was confirmed through ITS sequencing and a MoT3-specific molecular marker assay, with symptoms identical to those in the field developed on leaves and spikes inoculated with the isolate.

Genomic rearrangements generate hypervariable mini-chromosomes in host-specific isolates of the blast fungus

Thorsten Langner et al.

Summary: Supernumerary mini-chromosomes in the blast fungus M. oryzae are highly diverse and enriched in repetitive genetic elements, with lower gene density compared to core-chromosomes. These mini-chromosomes may emerge from structural rearrangements and segmental duplication of core-chromosomes, contributing to the adaptive evolution of the fungus. They contain virulence-related genes and are involved in large-scale genomic rearrangements, indicating their role in facilitating genome dynamics and potentially adaptive evolution.

Deciphering the role of microRNAs during Pi54 gene mediated Magnaporthe oryzae resistance response in rice

Kirti Arora et al.

Summary: The broad-spectrum resistance gene Pi54 in rice confers resistance against multiple isolates of Magnaporthe oryzae, with a variety of miRNAs identified to play a role in balancing the interaction between different defense response genes during Pi54 mediated resistance in the rice line Taipei309(Pi54). Additionally, the study suggests that the Pi54 gene mediated blast resistance in rice is influenced by multiple microRNAs regulating the immunity pathways, resulting in an incompatible host-pathogen interaction.

PTI-ETI crosstalk: an integrative view of plant immunity

Minhang Yuan et al.

Summary: Plants have innate immune responses to resist pathogen attacks, involving pattern-recognition receptors (PRRs) and nucleotide-binding domain leucine-rich repeat containing receptors (NLRs) that lead to pattern-triggered immunity (PTI) and effector-triggered immunity (ETI). Despite different activation mechanisms, PTI and ETI converge into similar downstream responses, with evidence of interactions and shared components between PRR-mediated and NLR-mediated signaling cascades. Future research on signal collaboration between PRR-initiated and NLR-initiated immunity will provide a more comprehensive understanding of the plant immune system.

Trans-kingdom RNAs and their fates in recipient cells: advances, utilization, and perspectives

Summary: The phenomenon and potential mechanisms of trans-kingdom RNA silencing (or RNA interference, RNAi) are currently one of the most exciting topics in science. Host-induced gene silencing (HIGS) based on trans-kingdom RNAi has great potential in creating crops resistant to various pests and pathogens. However, the lack of transformation technology in many crops limits the application of this technology, leading researchers to propose a possible alternative strategy for plant protection against pathogens without the need for crop genetic modification.
