Summary: Designing effective metallodrugs with amphiphilic nature is a promising approach for biomedical applications, as shown by the interaction of surfactant-ruthenium(II) complexes and precursor ruthenium(II) complexes with CT-DNA. The studies revealed that the surfactant-ruthenium(II) complexes bind to CT-DNA through hydrophobic interactions, while precursor ruthenium(II) complexes interact with CT-DNA through electrostatic interactions.

Novel polyamide amidine anthraquinone platinum(II) complexes: cytotoxicity, cellular accumulation, and fluorescence distributions in 2D and 3D cell culture models

Summary: In this study, aminoalkylamine substituted anthraquinones were peptide coupled to pyrrole lexitropsins and then converted into platinum(II) amidine complexes. Despite lower cellular accumulation compared to cisplatin, the platinum complexes exhibited cytotoxicities comparable to the standard drug. The platinum complexes with two or three pyrrole groups showed the lowest accumulation, but the three-pyrrole group complex demonstrated the highest activity, outperforming cisplatin in a specific cell line.

Self-Assembled Platinum Supramolecular Metallacycles Based on a Novel TADF Photosensitizer for Efficient Cancer Photochemotherapy

Summary: This study introduces a highly emissive supramolecular platinum triangle BTZPy-Pt based on a novel photosensitizer BTZPy with superior O-1(2) generation capability and high quantum yields. The combination of excellent phototoxicity of BTZPy and antitumor activity of the Pt center demonstrates a highly efficient anticancer performance towards HeLa cells. This research not only provides a blueprint for fabricating new types of photosensitizers, but also opens up possibilities for designing novel SCCs for efficient PDT.

Summary: A series of rhenium(I) polypyridine complexes with 2,4-dinitrophenylsulfonamide (DNPS) units were synthesized and characterized for their phosphorogenic bioimaging and photocytotoxic properties. The complexes exhibited weak luminescence upon photoexcitation due to photoinduced electron transfer, but showed enhanced emission and potent photocytotoxicity after reaction with glutathione (GSH) in living cells. Additionally, modification with a tosylamide unit enabled the complexes to selectively target the endoplasmic reticulum (ER) for bioimaging and photocytotoxic applications.

Summary: A family of six Ru(II) polypyridyl complexes with different lengths of alkyl chains have been synthesized and characterized, showing an influence on their photophysical and photochemical properties. The complexes with 21 carbon alkyl chains exhibited changes in emission properties and photosensitization abilities. The study also revealed that the complexes bind to DNA and demonstrated a length-dependent cellular uptake and cytotoxicity in cancer cells.

Luminescent naphthalimide-tagged ruthenium(ii)-arene complexes: cellular imaging, photocytotoxicity and transferrin binding

Summary: Two water-soluble piano-stool shaped ruthenium(ii)-arene complexes were designed as emissive photocytotoxic agents tagged with morpholine as the lysosome targeting moiety. They showed enhanced photo-cytotoxicity under low-power blue LED light irradiation, preferentially localized in lysosomes and nuclei, effectively targeting and damaging nuclear DNA for photodynamic therapy. Due to enhanced lipophilicity of one complex, it showed higher internalization into cancer cells, with possible binding through histidine residues and internalization via endocytosis. Luminescence properties were utilized to study cellular uptake mechanisms via endocytosis.

Fluorescent palladium(II) and platinum(II) NHC/1,2,3-triazole complexes: antiproliferative activity and selectivity against cancer cells

Summary: Fluorescent Pd(II) and Pt(II) complexes with 4-methylene-7-methoxycoumarin (MMC) and 2,6-diisopropylphenyl (Dipp) substituted NHC/1,2,3-triazole hybrid ligands were described, showing antiproliferative activity against cervix and breast cancer cells, with the tetradentate coordination mode being notably more effective. The Pd(s) complex with bidentate coordination mode was only active against HeLa, exhibiting high selectivity for cancer cells compared to healthy skin cells. The compound is believed to be located in late endosomes or lysosomes according to fluorescence microscopy.
