Summary: Diffuse intrinsic pontine glioma (DIPG) and other H3K27M-mutated diffuse midline gliomas (DMGs) are lethal pediatric tumors of the central nervous system. GD2-directed chimeric antigen receptor (CAR) T cells show promising preclinical efficacy. Four patients with H3K27M-mutated DIPG or spinal cord DMG were treated with GD2-CAR T cells and showed clinical and radiographic improvement.

Imipridones affect tumor bioenergetics and promote cell lineage differentiation in diffuse midline gliomas

Summary: The imipridone ONC201 and its derivative ONC206 significantly reduce cell viability in pediatric diffuse midline gliomas (DMGs) and prolong the survival of DMG PDX models. Both drugs impair tumor cell growth and survival by activating mitochondrial metabolism and the integrative stress response (ISR), and trigger cell lineage differentiation. These findings are of great importance for the treatment of pediatric DMGs.

Single-cell atlas of early human brain development highlights heterogeneity of human neuroepithelial cells and early radial glia

Summary: Through single-cell RNA sequencing, nine progenitor populations at the earliest stages of human brain development were identified, including a highly prevalent mesenchymal-like population that disappears once neurogenesis begins. Comparison with mouse progenitor populations revealed two clusters enriched in the early stages of human cortical development. Organoid systems displayed low fidelity to neuroepithelial and early radial glia cell types, but improved as neurogenesis progressed.

Summary: Single-cell RNA sequencing was used to examine the cell cycle states of expanding human neural stem cells (hNSCs), revealing a quiescent-like state in neuroepithelial-derived cells and gliomas. Neural G0 markers were enriched with quiescent NSC genes, and were significantly associated with less aggressive tumors and extended patient survival in gliomas. Knockout of genes associated with the Hippo/Yap and p53 pathways diminished Neural G0, resulting in faster G1 transit and down-regulation of quiescence-associated markers.
