Hexokinase 2 discerns a novel circulating tumor cell population associated with poor prognosis in lung cancer patients

Liu Yang et al.

Summary: This study demonstrates hexokinase-2 (HK2) as a metabolic function-associated marker for the detection of circulating tumor cells (CTCs) in non-small cell lung cancer (NSCLC) patients. The HK2(high)/CKneg CTCs exhibit metastasis and EGFR inhibitor resistance-related molecular signatures, and are selectively enriched in patients with EGFR(L858R) driver oncogene mutation. Patients with a larger number or proportion of HK2(high)/CKneg CTCs in the blood show poor therapy response and shorter progression-free survival.

Large Extracellular Vesicle Characterization and Association with Circulating Tumor Cells in Metastatic Castrate Resistant Prostate Cancer

Anna S. Gerdtsson et al.

Summary: Non-invasive liquid biopsies are a promising approach for tumor diagnosis and monitoring, with a focus on large extracellular vesicles (LEVs) which may provide informative sources for diagnosis and prognosis due to their increased presence in aggressive cancers. This study introduces a method for parallel identification and molecular characterization of LEVs and circulating tumor cells (CTCs), demonstrating that analysis of LEVs can enhance the sensitivity of liquid biopsy assays. The identification of LEVs in liquid biopsies offers potential for more comprehensive assessment of disease states for precise diagnosis and monitoring.

Circulating Tumor Cell Clusters Are Frequently Detected in Women with Early-Stage Breast Cancer

Carolina Reduzzi et al.

Summary: The majority of breast cancer-related deaths are caused by metastases, which are believed to originate from circulating tumor cells (CTCs), particularly CTC-clusters. Current conventional technology limits the early-stage analysis of CTCs, but a marker-independent blood filtration method has shown promise in detecting a high number of CTC-clusters in stage II-III breast cancer patients. This suggests that CTC-clusters may be an early event in the natural history of breast cancer, with marker-independent approaches improving their detection rates.

Profiling heterogenous sizes of circulating tumor microemboli to track therapeutic resistance and prognosis in advanced gastric cancer

Yang Chen et al.

Summary: Circulating tumor microemboli (CTM) aggregated by >= 2 circulating tumor cells (CTCs) exhibit higher migratory ability compared to single CTCs. The heterogeneous sizes of CTM in gastric cancer patients are associated with therapeutic resistance, with CTM containing 3-4 CTCs specifically linked to resistance to chemotherapy/targeted therapy and poor prognosis. Larger CTM clusters, >= 5 CTCs, are only detected intra-therapeutically and indicate liver metastasis in HER2(+) gastric cancer patients.

Summary: CTCs are cancer cells that can migrate through the circulatory system and cause metastasis. Liquid biopsy, especially that involving the collection of CTCs, is a non-invasive method to detect tumor cells in the circulatory system, allowing for earlier and easier detection than traditional tissue biopsies. It is crucial to develop accurate detection methods for further clinical applications due to the scarcity of CTCs in blood vessels.
