Sarecycline treatment for acne vulgaris: Rationale for weight-based dosing and limited impact of food intake on clinical efficacy

Ayman Grada et al.

Summary: The study investigated the impact of food intake on the absorption and clinical efficacy of sarecycline, a new oral narrow-spectrum third-generation tetracycline-class antibiotic approved for acne vulgaris treatment. Co-administration with high-fat food was found to decrease exposure by 21.7%, but had limited impact on clinical efficacy, allowing for weight-based dosing and flexibility in administration.

Global burden of bacterial antimicrobial resistance in 2019: a systematic analysis

Christopher J. L. Murray et al.

Summary: Antimicrobial resistance (AMR) poses a significant threat to global human health, and this study provides the most comprehensive estimates of AMR burden to date. By estimating deaths and disability-adjusted life-years (DALYs) associated with bacterial AMR in 204 countries and territories in 2019, the study highlights the impact of resistance and the leading pathogen-drug combinations contributing to it.

Sarecycline Demonstrated Reduced Activity Compared to Minocycline against Microbial Species Representing Human Gastrointestinal Microbiota

Mahmoud A. Ghannoum et al.

Summary: This study evaluated the impact of sarecycline on gut microbiota and found that it had reduced antimicrobial activity compared to minocycline against a wide range of microorganisms. This suggests that sarecycline is less disruptive to gut microbiota.

The Development of Third-Generation Tetracycline Antibiotics and New Perspectives

Aura Rusu et al.

Summary: The tetracycline antibiotic class has introduced new valuable members such as tigecycline, omadacycline, eravacycline, and sarecycline, which have excellent development potential. These modern tetracyclines have a broad antibacterial spectrum and are effective in treating conditions like acne. The class continues to be of interest to researchers for both antibiotic and other biological effects, with new derivatives being developed.

Increased prevalence of doxycycline low-susceptible Cutibacterium acnes isolated from acne patients in Japan caused by antimicrobial use and diversification of tetracycline resistance factors

Sae Aoki et al.

Summary: The study found changes in antimicrobial use and antimicrobial-resistant Cutibacterium acnes in acne patients following revisions in the Japanese acne treatment guidelines, particularly showing higher prevalence of low doxycycline-susceptible strains in patients with a history of oral tetracycline use. This highlights the importance of monitoring antimicrobial resistance patterns and the rapid dissemination of tetracycline resistance in C. acnes.

Antibacterial Mechanisms and Efficacy of Sarecycline in Animal Models of Infection and Inflammation

Christopher G. Bunick et al.

Summary: Sarecycline, a narrow-spectrum third-generation tetracycline-class antibiotic approved by the FDA for treating moderate-to-severe acne, exhibits activity against Gram-positive bacteria, particularly Staphylococcus aureus, with reduced efficacy against Gram-negative bacteria like Escherichia coli. Its unique C7 moiety interacts with bacterial ribosomes to block translation and prevent resistance, showing promising results in in vivo studies of infection and inflammation.
