Prostaglandin EP3 receptor signaling is required to prevent insulin hypersecretion and metabolic dysfunction in a non-obese mouse model o insulin resistance

Jaclyn A. Wisinski et al.

Summary: The study found that EP3 loss did not provide any diabetes protection to BTBR-Ob mice, but unexpectedly worsened insulin resistance and glucose tolerance in BTBR-lean mice. Additionally, it was discovered that BTBR-lean mice exhibited selective upregulation of constitutively active EP3 splice variants in their islets, leading to uncontrolled insulin hypersecretion.

Systemic Metabolic Alterations Correlate with Islet-Level Prostaglandin E2 Production and Signaling Mechanisms That Predict β-Cell Dysfunction in a Mouse Model of Type 2 Diabetes

Michael D. Schaid et al.

Summary: In the transition from beta-cell compensation to beta-cell failure in Type 2 diabetes (T2D), the role of Prostaglandin EP3 receptor (EP3) and its ligand prostaglandin E-2 (PGE(2)) has been demonstrated. By studying a rare genetic model of T2D in mice, researchers found improvements in EP3-mediated beta-cell dysfunction correlated with changes in metabolic parameters and inflammation markers.

Prolonged Low-Dose Dioxin Exposure Impairs Metabolic Adaptability to High-Fat Diet Feeding in Female but Not Male Mice

Summary: The study found that prolonged exposure to low-dose TCDD had minimal effects on glucose homeostasis and islet morphology in chow-fed male and female mice, but promoted maladaptive metabolic responses in HFD-fed females.

Rat prostaglandin EP3 receptor is highly promiscuous and is the sole prostanoid receptor family member that regulates INS-1 (832/3) cell glucose-stimulated insulin secretion

Harpreet K. Sandhu et al.

Summary: The study identifies EP3 as a key regulator of beta-cell function, with many agonists believed to be selective for other prostaglandin receptor family members also capable of activating EP3. Researchers need to be cautious when interpreting experimental results from cells or cell lines that express EP3.

Gsα-dependent signaling is required for postnatal establishment of a functional β-cell mass

Berta Serra-Navarro et al.

Summary: This study reveals the essential role of Gs protein in early postnatal beta-cell growth and maturation, partly mediated through its crosstalk with insulin signaling pathways. The findings suggest a close connection between these two pathways in postnatal beta cells, potentially implicating the use of cAMP-raising agents for promoting beta-cell regeneration and maturation in diabetes.

Pharmacological blockade of the EP3 prostaglandin E2 receptor in the setting of type 2 diabetes enhances β-cell proliferation and identity and relieves oxidative damage

Karin J. Bosma et al.

Summary: EP3 blockade promotes an increase in beta cell mass in db/db mice with type 2 diabetes, without affecting alpha cell proliferation. This effect may be mediated through the regulation of Nrf2, which plays a key role in protection against oxidative damage.

Regulation of Tissue Inflammation by 12-Lipoxygenases

Abhishek Kulkarni et al.

Summary: 12-LOX is a key lipid metabolizing enzyme that plays an important role in inflammatory diseases. Increased activity of 12-LOX can lead to tissue damage and enhanced migration of immune cells, while inhibition of this enzyme can protect against disease development and progression in animal models.

12-Lipoxygenase governs the innate immune pathogenesis of islet inflammation and autoimmune diabetes

Abhishek Kulkarni et al.

Summary: Macrophages and related myeloid cells play a crucial role in the early pathogenesis of T1D, with 12-LOX identified as a key enzyme promoting islet inflammation. Depletion of 12-LOX results in reduced macrophage infiltration, preservation of beta cell mass, and protection from T1D development in zebrafish and NOD mice models. The defect in myeloid cell migration caused by 12-LOX depletion is attributed to the loss of the chemokine receptor CXCR3. Transgenic expression of CXCR3 rescues the migratory defect.

Agonist-independent Gαz activity negatively regulates beta-cell compensation in a diet-induced obesity model of type 2 diabetes

Michael D. Schaid et al.

Summary: The inhibitory G protein alpha-subunit (G alpha(z)) plays a crucial role in modulating beta-cell function and offers protection against high-fat diet-induced diabetes. The altered gene expression pattern in islets from HFD-fed G alpha(z) KO mice compared to WT mice suggests a secretion-specific mechanism is involved in this protection. Additionally, G alpha(z) couples with the prostaglandin EP3 receptor in pancreatic beta cells to regulate cAMP production and downstream beta-cell function.

Human Islet Expression Levels of Prostaglandin E2 Synthetic Enzymes, But Not Prostaglandin EP3 Receptor, Are Positively Correlated with Markers of β-Cell Function and Mass in Nondiabetic Obesity

Nathan A. Truchan et al.

Summary: The production and signaling pathway of PGE(2) are associated with glycemic control, obesity, and systemic inflammation in both nondiabetic and type 2 diabetes subjects. Levels of IL-6 and COX-2 mRNA in islets show positive correlation with donor BMI, while IL-6 expression is strongly related to the expression of other PGE 2 synthetic pathway genes.

Prolonged Low-Dose Dioxin Exposure Impairs Metabolic Adaptability to High-Fat Diet Feeding in Female but Not Male Mice

Geronimo Matteo et al.

Summary: Our study found that prolonged low-dose TCDD exposure has minimal effects on glucose homeostasis and islet morphology in chow-fed male and female mice, but promotes maladaptive metabolic responses in HFD-fed females. Females exposed to TCDD showed accelerated onset of HFD-induced hyperglycemia and impaired glucose-induced plasma insulin levels, with changes in islet area and beta cell percentages. TempO-Seq analysis suggested abnormal changes to endocrine and metabolic pathways in female TCDDHFD islets.
