HMDB 5.0: the Human Metabolome Database for 2022

David S. Wishart et al.

Summary: The Human Metabolome Database (HMDB) has been providing comprehensive information about human metabolites since 2007, and has undergone significant improvements and upgrades in its latest update, HMDB 5.0. These improvements include an increase in the number of metabolite entries, enhancements to metabolite descriptions, new visualization tools, and more accurately predicted spectral data sets. These upgrades are aimed at improving the usability and potential applications of the HMDB in various fields, including human metabolomics, exposomics, lipidomics, nutritional science, biochemistry, and clinical chemistry.

Genome-wide association studies of metabolites in Finnish men identify disease-relevant loci

Xianyong Yin et al.

Summary: The Finnish population has a high frequency of genetic variants that are rare in other populations. This study identifies new genetic loci associated with 1391 circulating metabolites in Finnish men, highlighting the potential of metabolite genetic associations in understanding disease mechanisms. The results reveal novel genes and mechanisms related to diseases and disease-related traits.

Titin-truncating mutations associated with dilated cardiomyopathy alter length-dependent activation and its modulation via phosphorylation

Petr G. Vikhorev et al.

Summary: This study investigates the effects of truncating mutations in the titin gene (TTN) on the function and characteristics of myofibrils. The researchers found that these mutations lead to decreased length-dependent activation and increased elasticity of myofibrils. They also observed decreased phosphorylation levels of TnI and MyBP-C in DCM patients with TTN-truncating variants.

Epidemiology of the inherited cardiomyopathies

William J. McKenna et al.

Summary: Estimates of the incidence and prevalence of inherited cardiomyopathies are challenging due to incomplete and late-onset disease expression, highly variable penetrance within families, and overlapping phenotypes. Current global estimates for hypertrophic, dilated, and arrhythmogenic cardiomyopathies may be conservative.

The Time Has Come to Explore Plasma Biomarkers in Genetic Cardiomyopathies

Nienke M. Stege et al.

Summary: Screening for pathogenic variants has become common practice for patients with genetic cardiomyopathies, allowing early detection of disease onset. Monitoring relies on a combination of plasma biomarkers, imaging techniques, and electrocardiography. Further research is needed to explore the potential of biomarkers in distinguishing primary cardiac disease from secondary organ dysfunction.

The Echocardiographic Parameters of Systolic Function Are Associated with Specific Metabolomic Fingerprints in Obstructive and Non-Obstructive Hypertrophic Cardiomyopathy

Martino Deidda et al.

Summary: The study aimed to evaluate if metabolomics combined with echocardiography could identify pathophysiological differences between obstructive and non-obstructive hypertrophic cardiomyopathy. Results revealed distinct differences in echocardiographic parameters and metabolomic profiles between the two phenotypes, with OHCM patients showing better contractility and specific metabolic activities associated with certain metabolites.
