NMR resonance assignments for a docking domain pair with an attached thiolation domain from the PAX peptide-producing NRPS from Xenorhabdus cabanillasii

Jonas Watzel et al.

Summary: Non-ribosomal peptide synthetases (NRPSs) are large multi-enzyme machineries that synthesize important natural products from amino acids. Specialized NRPS modules interact in a defined manner for peptide synthesis. The binding specificity of docking domains is crucial for the correct assembly of multi-protein NRPS systems.

New prodrugs and analogs of the phenazine 5,10-dioxide natural products iodinin and myxin promote selective cytotoxicity towards human acute myeloid leukemia cells

Summary: This study focuses on optimizing lead compounds in phenazine 5,10-dioxides and highlights the importance of carbamate side chains as the optimal phenol-attached group. Derivatives without oxygen-based substituents in the 6th position of the phenazine skeleton retained potency when alkyl or carbamate side chains were attached to the phenol in position 1. Additionally, 7,8-dihalogenated and 7,8-dimethylated analogs of 1-hydroxyphenazine 5,10-dioxide showed increased cytotoxic potency in MOLM-13 cells compared to other compounds studied.
