Reverse Transcriptase: From Transcriptomics to Genome Editing

Samara Martin-Alonso et al.

Summary: RTs are enzymes that can generate cDNA from RNA, widely used for detecting RNAs and cloning expressed genes. Improved by protein engineering, RTs are key elements in next-generation sequencing techniques and have shown potential for manipulating eukaryotic genomes.

Test sensitivity is secondary to frequency and turnaround time for COVID-19 screening

Daniel B. Larremore et al.

Summary: The COVID-19 pandemic has led to a public health crisis due to the spread of SARS-CoV-2 through presymptomatic, symptomatic, and asymptomatic individuals. Effective population screening for the virus is essential for reopening societies and controlling its spread, with testing frequency and reporting speed playing crucial roles in its success. Test sensitivity is only marginally important compared to these factors.

Clinical Assessment and Validation of a Rapid and Sensitive SARS-CoV-2 Test Using Reverse Transcription Loop-Mediated Isothermal Amplification Without the Need for RNA Extraction

Melis N. Anahtar et al.

Summary: In response to the urgent need for expanded COVID-19 testing, a simple and inexpensive RT-LAMP test was developed to detect SARS-CoV-2 in 40 minutes. By incorporating sample processing steps, the sensitivity and specificity of the assay were significantly improved, making it a promising option for increasing testing capacity in resource-limited settings.

Testing at scale during the COVID-19 pandemic

Tim R. Mercer et al.

Summary: The assembly and publication of the SARS-CoV-2 genome in January 2020 allowed for immediate development of tests and led to the largest global testing programme in history. Despite efforts to control the spread, widespread community transmission of the virus has become entrenched in many countries, necessitating population-scale testing to identify and isolate infected individuals. Testing plays a crucial role in the response to the COVID-19 pandemic and is likely to continue to be vital in public health.

Low saliva pH can yield false positives results in simple RT-LAMP-based SARS-CoV-2 diagnostic tests

Cristina Uribe-Alvarez et al.

Summary: Efficient diagnosis of infectious diseases such as COVID-19 is crucial for timely treatment. While RT-qPCR tests are ideal in hospital environments, cheaper and simpler testing methods are needed for mass testing. This study found that using nasopharyngeal swabs in VTM and saliva samples in an isothermal LAMP-based assay can provide reliable and low-cost testing for SARS-CoV-2 infection.

High-Surety Isothermal Amplification and Detection of SARS-CoV-2

Sanchita Bhadra et al.

Summary: Isothermal nucleic acid amplification tests, like loop-mediated isothermal amplification, offer a low-cost option for rapid visual detection, but false positive and negative results should be considered. Multiplex assays with logical signal readout and sequence-specific probes can help reduce the risk of misinterpretation.

Asymptomatic SARS-CoV-2 infection: A systematic review and meta-analysis

Pratha Sah et al.

Summary: Quantification of asymptomatic infections is crucial for effective public health responses to the COVID-19 pandemic. Analyzing over 350 studies, it was estimated that 35.1% of infections never developed clinical symptoms and were truly asymptomatic. At the time of testing, 42.8% of cases showed no symptoms, including both asymptomatic and presymptomatic infections.

Estimating infectiousness throughout SARS-CoV-2 infection course

Terry C. Jones et al.

Summary: In a study of SARS-CoV-2 cases in Germany, it was found that younger subjects had lower viral loads but higher cell culture replication probabilities compared to older subjects. Additionally, 8% of subjects had viral loads above 10(9) copies per swab, with a third of them being presymptomatic, asymptomatic, or mildly symptomatic individuals with a mean age of 37.6 years. The B.1.1.7 lineage infections had a higher viral load and cell culture replication probability than non-B.1.1.7 subjects.

Diagnostic accuracy of rapid antigen tests in asymptomatic and presymptomatic close contacts of individuals with confirmed SARS-CoV-2 infection: cross sectional study

Ewoud Schuit et al.

Summary: The diagnostic test accuracy of two rapid antigen tests in asymptomatic and presymptomatic close contacts of people with SARS-CoV-2 infection on day 5 after exposure was more than 60%, increasing to more than 85% after a viral load cut-off was applied as a proxy for infectiousness.

Accuracy of novel antigen rapid diagnostics for SARS-CoV-2: A living systematic review and meta-analysis

Lukas E. Brummer et al.

Summary: A systematic review and meta-analysis was conducted to evaluate the clinical accuracy of commercially available Ag-RDTs for SARS-CoV-2. The study found that Ag-RDTs can detect the vast majority of infected individuals in the early phase of the disease and those with high viral load, making them valuable diagnostic tools. However, concerns were raised regarding bias across datasets and the need for standardization in conducting and reporting clinical accuracy studies for improved comparability and data utilization.

Diagnostic performance of a colorimetric RT -LAMP for the identification of SARS-CoV-2: A multicenter prospective clinical evaluation in sub-Saharan Africa

Marycelin Mandu Baba et al.

Summary: This multicenter prospective observational study evaluated the performance of RT-LAMP in resource-limited settings in four African countries and Italy. The study found that RT-LAMP demonstrated high specificity and positive predictive value, especially with medium-high viral loads, making it a reliable assay for timely management and prevention of COVID-19 without compromising quality.
