Identification of (Z)-2-benzylidene-dihydroimidazothiazolone derivatives as tyrosinase inhibitors: Anti-melanogenic effects and in silico studies

Summary: In this study, a series of tyrosinase inhibitors based on the structure of MHY773 were designed and evaluated. Three DHIT derivatives, 1a, 1b, and 1f, showed stronger inhibitory effects on mushroom tyrosinase than kojic acid. Compound 1b demonstrated superior competitive inhibition against mushroom tyrosinase compared to kojic acid and MHY773. Furthermore, compounds 1b and 1f exhibited potent inhibitory effects on cellular tyrosinase activity and melanin production.

Anti-Tyrosinase Compounds from the Deep-Sea-Derived Actinomycete Georgenia sp. 40DY180

Summary: In this study, a method called assay-guided approach was used to discover compounds that modulate tyrosinase activity from marine actinomycete. A new compound, georgenione A (1), along with two previously known compounds, 5-(4'-hydroxybenzyl)hydantoin (2) and cyclo(Trp-Gly) (3), were isolated from actinomycete Georgenia sp. 40DY180. Compound 2 showed strong anti-tyrosinase activity, comparable to the commercially used positive controls kojic acid and arbutin.

Summary: A series of novel 1,2,4-triazole compounds were synthesized and their structures were characterized through spectral methods. The synthesized derivatives showed good inhibitory activity against mushroom tyrosinase, with N-(4-fluorophenyl)-2-(5-(2-fluorophenyl)-4-(4-fluorophenyl)-4H-1,2,4-triazol-3-ylthio) acetamide exhibiting the most prominent activity. Molecular docking studies revealed the interaction profile of these compounds at the active site of tyrosinase.
