Regioselective and Stereoselective Epoxidation of n-3 and n-6 Fatty Acids by Fungal Peroxygenases

Summary: Research on epoxide metabolites from n-3 and n-6 polyunsaturated fatty acids has shown that fungal unspecific peroxygenases can convert these fatty acids into mono-epoxides with high regio- and enantioselectivity. This study highlights the potential synthetic utility of fungal peroxygenases in fatty acid epoxidation.

Pilot-Scale Production of Peroxygenase from Agrocybe aegerita

Summary: The pilot-scale production of the peroxygenase from Agrocybe aegerita (rAaeUPO) is demonstrated, resulting in a final concentration of 0.29 g L-1 after 6 days of fermentation. However, only 170 g of the enzyme has been isolated due to nonoptimized downstream processing. The enzyme preparation showed preparative usefulness at a semipreparative scale for the stereoselective hydroxylation of ethyl benzene, achieving more than 340,000 catalytic turnovers and producing over 5 g L-1 (R)-1-phenyl ethanol in 7 days.
