Intracellular ion changes induced by the exposure to beta-amyloid can be explained by the formation of channels in the lysosomal membranes

Dmitry Zaretsky et al.

Summary: This manuscript reevaluates previous data on beta-amyloid-induced changes of intracellular ions concentrations published by Abramov et al. (2003, 2004). While their observations support the amyloid channel theory, a closer look reveals facts that cannot be explained by channel formation in the plasma membrane. The recently proposed amyloid degradation toxicity hypothesis offers an interpretation to these facts by considering channels formed in lysosomal membranes.

Alzheimer's disease: unraveling APOE4 binding to amyloid-beta peptide and lipids with molecular dynamics and quantum mechanics

Jean-Numa Gillet

Summary: The study unravels the binding mechanisms of APOE4 associated with Alzheimer's disease, showing that when bound to the amyloid-beta peptide, APOE4 undergoes misfolding and increased deposition, potentially leading to the disease. Immunotherapies targeting APOE4 are promising for drug design, but the focus should be on non-lipidated APOE4.

Decreased pH in the aging brain and Alzheimer's disease

Yann Decker et al.

Summary: The study compared pH levels in the human brain and cerebrospinal fluid (CSF) in postmortem control and Alzheimer's disease cases using publicly available data sets. Results showed a reduction in pH with aging and a further decrease in Alzheimer's disease cases. In vivo experiments demonstrated that low pH CSF infusion increased amyloid beta plaque load, while in vitro studies showed that mild acidosis reduced inflammation and amyloid beta uptake by microglia. These findings suggest that brain acidosis may play a role in Alzheimer's disease pathophysiology.

Summary: This study focused on the structural properties, vibrational analysis, and reactivity of the novel insecticide flonicamid. Eight minimum energy conformers were identified and analyzed, with good correlation between theoretical and experimental data. Docking analysis suggested potential carcinogenic effects that should be experimentally evaluated.

Endo-lysosomal Aβ concentration and pH trigger formation of Aβ oligomers that potently induce Tau missorting

Marie P. Schutzmann et al.

Summary: The formation of A beta oligomers (A beta Os) is significantly accelerated in endo-lysosomal pH conditions, leading to extensive A beta O formation under physiological conditions, which triggers Alzheimer's disease-related pathologies. Exploiting a dimeric variant of A beta, the authors demonstrate that this model construct is relevant for studying pathogenic A beta O formation and its effects on Tau missorting and impaired neuronal activity.

Summary: Alzheimer's disease has a significant impact on individuals and society as a whole, with a large number of patients in the United States and a potential for continued growth in the future. The value of unpaid caregivers has increased despite a decline in their numbers, and healthcare costs are on the rise. Racial and ethnic disparities in healthcare persist, highlighting the need for greater diversity in dementia care workforce and clinical trials.
