A CRISPR/Cas9-Mediated, Homology-Independent Tool Developed for Targeted Genome Integration in Yarrowia lipolytica

Summary: This study developed a homology-independent targeted genome integration tool mediated by CRISPR/Cas9 in Yarrowia lipolytica. The tool does not require homologous templates, and can be used to rapidly verify genetic elements and iteratively integrate multiple-gene pathways in Y. lipolytica. This tool may serve as a potential supplement to current HR-dependent genome-editing methods for eukaryotes.

A Golden-Gate Based Cloning Toolkit to Build Violacein Pathway Libraries in Yarrowia lipolytica

Summary: Violacein, a naturally occurring anticancer therapeutic compound, was produced in Y. lipolytica using a library of strains constructed with Golden Gate assembly method. Gene expression analysis revealed that strong expression of VioB, VioC, and VioD favored violacein production, while weak expression of VioD was associated with high deoxyviolacein production. By optimizing culture conditions, violacein reached a maximum of 70.04 mg/L with 5.28 mg/L of deoxyviolacein in shake flasks.

Production of (-)-α-bisabolol in metabolically engineered Saccharomyces cerevisiae

Summary: Metabolic engineering strategies were employed to produce (-)-alpha-bisabolol in Saccharomyces cerevisiae, resulting in increased production through optimized gene expression, intensified mevalonate pathway, and enhanced acetyl-CoA pool. The engineered strain achieved significantly higher yield and productivity of (-)-alpha-bisabolol in fed-batch fermentation compared to the initial strain in batch culture, indicating the potential for sustainable production of sesquiterpenes in engineered S. cerevisiae.
