Erosive hand osteoarthritis: latest findings and outlook

Adiponectin in Osteoarthritis: Pathophysiology, Relationship with Obesity and Presumptive Diagnostic Biomarker Potential

Summary: Osteoarthritis (OA) is a multifactorial degenerative disease that is irreversible and induced by age and obesity. Currently, the pathophysiology and etiology of OA remain under-investigated, and the available treatment relies mainly on total joint arthroplasty, which is costly. Recent research has focused on evaluating metabolically active mediators secreted by adipose tissue, with a particular interest in adiponectin due to its association with OA. However, studies on the role of adiponectin in OA development have produced conflicting results. This review critically examines the roles of adiponectin in OA pathophysiology, the link between obesity, adiponectin expression, and OA progression, as well as its potential as a biomarker for improved diagnosis of this degenerative disorder.

Leptin in Osteoarthritis and Rheumatoid Arthritis: Player or Bystander?

Summary: White adipose tissue is a specialized tissue involved in lipid synthesis and storage. It serves as an active organ that secretes hormones and cytokines called adipokines, with leptin being a key molecule. Leptin not only regulates energy balance and food intake, but also plays roles in reproduction, glucose metabolism, hematopoiesis, and immune system interactions. It is involved in innate and adaptive immunity and contributes to the pathogenesis of chronic inflammation and immune-related diseases like osteoarthritis and rheumatoid arthritis. This review summarizes recent findings on the involvement of leptin in immunity and inflammatory responses in these diseases.

Effects of the Leptin-Mediated MAPK/ERK Signaling Pathway on Collagen II Expression in Knee Cartilage of Newborn Male Mice from Obese Maternal Offspring

Innovative therapeutic approaches are needed to effectively repair and restore damaged synovial joints in this population.

JUNB-FBXO21-ERK axis promotes cartilage degeneration in osteoarthritis by inhibiting autophagy

Chondrocytes from osteoarthritic cartilage of obese patients show altered adiponectin receptors expression and response to adiponectin

Summary: This study revealed that adiponectin receptor ADIPOR1 is the most abundant in primary OA chondrocytes, with differences in expression between lean and obese patients. Adiponectin was found to induce the expression of MCP-1, IL-6, and MMP-1 genes in all OA patients' chondrocytes, while the regulation of VCAM-1 and MMP-2 by adiponectin varied depending on the patient's body mass index. These findings suggest that adiponectin and ADIPOR1 may play crucial roles in the pathogenesis of cartilage degeneration in obese individuals with OA.

Vaspin deficiency failed to promote the proliferation of BMSCs in osteoarthritis
