Immunological considerations and challenges for regenerative cellular therapies

Sandra Petrus-Reurer et al.

Summary: The article discusses the roles of both innate and adaptive immune responses in allogeneic graft rejection, as well as how these responses are determined by the cellular therapy being studied. It also describes the range of in vitro and in vivo approaches used to examine the immunogenicity of cellular therapies and considers potential confounders and strategies that can be employed to ameliorate immune rejection.

Mitomycin-Ctreatment during differentiation of induced pluripotent stem cell-derived dopamine neurons reduces proliferation without compromising survival or function in vivo

Benjamin M. Hiller et al.

Summary: This study presents effective non-genetic methods to restrain cell proliferation when generating dopamine neurons for Parkinson's disease transplantation, suggesting that the use of MMC can successfully reduce proliferation without compromising long-term survival and function of the grafts.

Mesenchymal stem/stromal cells: Developmental origin, tumorigenesis and translational cancer therapeutics

Chenghai Li et al.

Summary: While a large body of research has shown the dual role of MSCs in tumor growth and inhibition, studies on the contribution of MSCs to tumorigenesis are rare. MSCs can increase cancer stem cell population, generate cancer precursors, and induce epithelial-mesenchymal transition. Understanding the developmental origin and tissue-resident native MSCs is crucial for grasping MSCs' contribution to cancer cell origin.

Review: Tissue Engineering of Small-Diameter Vascular Grafts and Their In Vivo Evaluation in Large Animals and Humans

Shu Fang et al.

Summary: The current research on small-diameter tissue-engineered vascular grafts (SD-TEVGs) focuses on improving design to address issues like mechanical failure, thrombogenicity, and intimal hyperplasia that have limited success so far. Synthetic polymer-based SD-TEVGs have shown limited success in large-animal models, while natural polymer and cell-secreted ECM-based SD-TEVGs are still facing challenges in terms of strength and thrombogenicity. Further exploration of allogeneic native ECM-based SD-TEVGs, in vitro-developed hybrid SD-TEVGs with allogeneic banked human cells or isolated autologous stem cells, and in-body tissue architecture (IBTA)-based SD-TEVGs appear promising for the future due to their dimension, mechanical strength, biocompatibility, and availability.

Patches of different types for carotid patch angioplasty (Review)

Saritphat Orrapin et al.

Summary: The safety and efficacy of different types of patch materials used in carotid patch angioplasty were evaluated. Most studies suggested little to no difference between autologous vein and synthetic materials in perioperative and long-term ipsilateral stroke outcomes.
